For producing feeding proteins and feeding multi-enzyme, fermentation with distiller's grain (such as yellow wine lees and beer lees) and farm produce dregs (such as bean cake and bran)and corn cob hydrolysis was studied. The aim is to deal with the environment problem induced by distiller's grain and reuse the cellulose resource.The key point is producer. The initial strain, mainly producing protease and cellulase, was mutagenized by heat treatment and 60Co radial. Two mutants were gained. One is M-41 producing protease (2380U/g dry mass) which was enhanced by 37%, and the other one is M-462 producing cellulase (1068 U/g dry mass) which was enhanced by 45%. Then fermentation with these two strains was studied. And the preferable fermentation technics was that (g/L) bean cake 222, bran 500, urea 5, (NH4)2SO4 5, KH2PO4 8, MgSO4·7H2O 0.4, dry mass: water equal 1:1.2, pH5.0, at 30℃was cultivated for 40h. The protease and cellulase of product were 3135 U/g dry mass and 1543 U/g dry mass respectively.Enzyme property was also studied. It is found that, to protease, the optimum digestion temperature was 45℃,when digestion was carried at 40℃, the better time was1h. When substrate concentration was lower 0.8%, enzyme reaction velocity was direct ratio to substrate concentration. When substrate concentration was above 1.0%, enzyme reaction velocity increased little and enzyme was saturated. The optimum temperature arrange of enzyme reaction was 40~45℃, enzyme activity could hold 80% between 30~40℃. And the optimum pH arrange of enzyme reaction was 6.2~7.2,enzyme activity kept stable at pH6.8~7.2. To cellulas, the optimum digestion temperature was 50℃. When digestion was carried at 40℃, the better time was 2h. When substrate concentration was lower 1.0%, enzyme reaction velocity was direct ratio to substrate concentration. When substrate concentration was above 1.2%, enzyme was saturated. The optimum temperature arrange of enzyme reaction was 35~45℃, enzyme activity could hold 80% between 30~60℃. And the optimum pH arrange of enzyme reaction was 2.2~5.0,enzyme activity kept stable at pH3.0~6.0. The solid-state fermentation was carried with blending Aspergillus oryzae and yeast to product protein-enriched feed including enzyme.
