Abstract/Summary:
Mastitis is one of the main diseases harmful to the development of diary industry, which is also a serious zoonosis. Clinical mastitis incidence is 2%-3%, which accounts for 20%-25% of the total incidence in the cow disease, while the subclinical mastitis incidence is 38%-62%. According to the investigation of United States, this disease has caused a loss of 1.3 billion each year. The incidence in China is higher as a result of the separating breed, poor management conditions, incomplete detecting techniques and so on. So it is quite necessary to detect the mastitis to assure the health of consumers and the quality of dairy products. Normal mastitis-detecting techniques use as microscopical counts, CMT, LMT etc. researches have been done about pathogenic microbe of mastitis at present, most of them being bacterial infected, Streptococcus and staphylococcus aureus are world wide spreading. Basing on the MTT theory, our experiment developed a MTT-detecting method of mastitis and put it into the research of antibiotic susceptibility test.1. In order to develop MTT-detecting method of mastitis, the effect of milk sample dilute range,lipid,terminative reagent and different culture times on colour change were determined, select the optional detecting conditions were milk sample dilute twice directly without getting rid of lipid, terminative buffer was is SDS-DMF,and best culture time was 8 h.2. Comparing the method established in our experiment with Bacterial-Counts and LMT. The results showed it has good consistency with the two methods above. Compared to Bacterial-Count it showed the advantages below:â‘ Better specificity;â‘¡Rapidness and veracity;â‘¢higher sensitivity;â‘£Better consistency. And it is fit to detect mastitis in prophase comparing with LMT. The lowest microbial content detect limit in milk sample is 800 cfu?mL-1, while under 95 cfu?mL-1 being undetectable.According to the investigation of mastitis in the teach-base of North West Sci-Tech university of A&F, the incidence of positive mastitis by MTT was 40.91% while 32.95% by LMT.
