The experiment were conducted of the effect of E2 ,P4,OT and PGF2αon proliferation of Peripheral Blood Mononuclear cell(PBMC) in swine at different concentrations, and the effect of these hormones on secreting IL-4 of PBMC by MTT assay ,in order to detect the roles and there mechanism of these hormones on immune system and to illuminate the relationship between endocrine and immune system at different physiological periods. The results are following.1. The Results of orthogonal test showed that the stimulating activity of four kinds of hormones on PBMC in different concentrations is distinguished. The effect of PGF2αis the strongest of all, OT is the next, and then is E2, P4. The effect is the strongest when E2 is 10 pg/mL, P4 is 1ng/mL,OT is 0.2pg/mL,and PGF2αis 1ng/mL. The effect of hormones on immune system are vary when the concentration are different.2. Four hormones stimulate the PBMC singly, which proliferation activity is higher than co-stimulation of the hormones and PHA-P or that of PHA-P. Meanwhile, when the hormones are co-stimulating with PHA-P, except P4, the hormones concentration of the peak value are different with the hormone used single. When hormones stimulate the PBMC singly, the transformation is most significantly at the concentration of E2 10 pg/mL ,P4 1 ng/mL,OT 0.2 pg/mL ,PGF2α1 ng/mL separately. While the hormones co-stimulation with PHA-P ,the concentration is E2 100 ng/mL, P4 1 ng/mL,OT 2 ng/mL ,PGF2α100 ng/mL separately. It suggested that the reproduction hormones have different mechanism with PHA-P and they can active different kinds of lymphocytes, which shows the complexity and specificity of hormones in stimulating the lymphocytes.3. Four hormones can all stimulate the PBMC to secrete IL-4 which is dose-dependent, but the rules were distinguished. E2 and P4 at100ng/mL, the content of IL-4 is the maximum, but it would degrade when super- or infra- concentration. With the increase of OT concentration the content of IL-4 is degrade. The role of PGF2αwas not obviously.
