Adipose tissue has endocrinic function which can secrete many kinds of adipokines. These cytokines affect the metabolism of adipose tissue and the whole body of the organism. The distinct characteristic of cytokines working fashion is strictly regulated by time and space. There are at least three kinds of protein families involved in the negatively regulating the cytokines'signaling: SOCS, PIAS and SHP-1. Among them, the study of the SOCS family was relatively extensive. SOCS-3 is one of the main members of the SOCS family. Many cytokines'signaling such as Leptin, insulin and IGF-â… was regulated by SOCS-3 both in adipose tissue and muscle tissue. Therefore, in this study, we investigated the expression pattern of SOCS-3 mRNA in pig adipose and muscle tissue and tried to explore its regulation mechanism. The main results were summarized as following:1. Successfully cloned the pig SOCS-3 cDNA gene, and the Genbank accession number is DQ644577. Homologous alignment analysis showed that the porcine SOCS-3 gene has 92%,88%and 88%homology to that of human, rat and mouse respectively. Further CDD analysis showed that the SOCS-3 protein which deduced from the cloned sequence has the SH2 domain and the SOCS-box which are the basic domains of SOCS family.2. The tissue distribution analysis showed that SOCS-3 transcript was positively detected in heart, liver, spleen, lung, kidney, muscle, subcutaneous adipose and visceral adipose. Among them, the lung and spleen had relatively higher SOCS-3 mRNA expression, while liver and visceral had lower SOCS-3 mRNA expression. Interestingly, the expression of SOCS-3 mRNA was significantly different between subcutaneous adipose and visceral adipose tissue. At the same time, SOCS-3 mRNA was positively determined in skeletal muscle. These results indicated that SOCS-3 may play important role in the different fat deposition ability between subcutaneous adipose and visceral adipose tissue. In addition, the function of SOCS-3 in muscle tissue also an attractive target for Chinese pig breeding scientists.3. Compared to Largewhite pigs, the mRNA expression of SOCS-3 in subcutaneous adipose and muscle tissue of Bamei pigs was increased during aging.
