Heilongjiang is one of the main potato breeding and multiplication base in china that the speed of breeding seedlings in vitro directly influence the developing of China potato industry. As basic stock of potato breeding and multiplication, to obtain vigorous seedlings in vitro is a crucial step. How to obtain vigorous seedlings with health roots, thick stems and more functional nodes would be beneficial for inducing microtubers. It is also a practical mean to boost popularization of new varieties.6 new varieties released by Northeast Agricultural University, Daxinganling Institute of Agricultural Sciences and Keshan Institute of Potato were applied to this test that are different fried chip ,fried slice and high content varieties .Those will become special potato processing varieties that have important function to potato processing industry.In order to establish culture system of vigorous seedlings and to speed up popularization , 6 different genotype varieties were researched by culture conditions and components of medium were optimized Potato with low cost and simple manipulation was applied to theoretics thereunder for large scale factory production.1. Seedling height, stem thick, number of leaves, number of roots, and length of roots were studied by means of double-factor complete random design. The result of variance analysis showed that significant differences existed in all characteristics among varieties, which meant genetic background among varieties tested were different, and separate medium should be adopted for various varieties.2. In order to make optimum mediums for each variety, some elements were adjusted based on performance of seedling height, stem thick, number of leaves, number of roots and length or roots in the orthogonal design. The results indicated that the optimum medium for Zhongda No.1 and Kexin No.16 was MS plus NAA 0.1mg/L,6-BA 0.1mg/Lå’ŒB9 20mg/L. The optimum medium for NEA303, Kexin No.4, Kexin No.15 was MS plus NAA 0.2mg/L,6-BA 0.1mg/Lå’ŒB9 20mg/L. The optimum medium for Kexin No.17 was MS plus NAA 0.2mg/L,6-BA 0.1mg/Lå’ŒB9 10mg/L.3. The result of dynamic analysis on seedling height, stem thick and number of functional nodes in consecutive four weeks showed that Kexin No.17 and Kexin No.16 had thick stems, more functional nodes and higher rate for multiplication. From the second week of culture in vitro, seedling height of Zhongda No.1 was higher than others significantly, and only three weeks are needed for maturing seedlings.
