Cucumber is one of important vegetables, and have important relation to people life. With the improvement of living level, we request higher quality. While, physiognomic quality is direct factor of purchasing desire. For these years, cucumber varieties of European type which have green and uniform pericarq, smooth pericarq, short bar shape, no or extremely short neck are popular. Now, most kinds of varieties depend on import. Therefore, in order to satisfy the demand of domestic market and need of export, we need to breed the cucumber varieties of European type.In order to select the cucumber material which are short neck accurately during the seeding period, rise the select efficiency about cucumber appearance character , shorten the period of breeding , quicken the course of cucumber quality breeding in our country . The statistical analysis population in our experiment is F1 and F2 population from crosses whose mother(short neck) is cucumber varieties Z3 coming from Dutch and father (long neck)is Dongnong 129 . The experiment use the method of bulk segregant analysis (BSA) and simple sequence repeat (SSR) to locate the QTL of the fruit neck length. The main result were as follows:1. Use F2 population which is come from 129×Z3 as mapping population. We get 5 pairs of SSR primers which are co-relation with the character about fruit neck by selected 86 pairs SSR primers with parent and DNA pool. The result was verified with 75 individuas in F2 population. Analyising with MAPMAKER/EXP version 3.0b CSWGATT01B, CSWGATT01C, CSCT335 and CSWGATT01A were located in one linkage group.2. Having detected the gauss distribution of fruit length and fruit neck character, we found the bias angle abs.v. of fruit length and fruit neck were smaller than 0.5. Bassed on this we reach a conclusion that these two character are accord with gauss distribution.3. Analys to QTL preliminary location we get approximate position of one QTL about fruit neck. Qchl 1 was located in the CSWGATT01A-CSWGATT01B linkage group, the concerat position is between CSWGATT01B and CSWGATT01C, the genetic distance from which Qchl 1 to both sides are 3.4 cM and 18.0 cM (LOD is 3.48). The accountable variability is 18.49%, additive is 0.84.4. The result of 30 germplasm identification is according with field performance. Based on this we can make a initiative conclusion that is the SSR market result which is about cucumber fruit neck character QTL position is correct.
