In this study, indirect enzymelinket immunosorbent assay(IELISA), immunohistochemistry staining , histochemistry staining was used to discover the changes of the contents both IgS antibody-producing cells and the number of CD4+, CD8+ lymphocyte subpopulation, TANAE+ cells in caecal tonsils, gland of Harder, Peyer's patch , the changes of the content of Igs in local humor–tear, trachea fluid, bile, intestinal fluid of 7-day-old and 21-day-old chicks infected with AIV, in order to reveal mechanism of immunity and provide scientific referential basis for the production of vaccine of avian influenza. The results were as follows: the content of the Igs intear, trachea fluid, bile, intestinal fluid and the number of IgA,IgG,IgM in caecal tonsils, gland of Harder, Peyer's patch of the chicks infected with AIV were decreased rapidly compared with controls 1 ~3 day post infection (PI) in different degree. The level of IgS and correspondly the numberof antibody-producing cells began to ascend 4~5 day PI, and were higher than controls continuely.close to controls at 21-day-old.in the early days, The level of IgS and the numberof antibody-producing cells was decreased rapidly and exhaustion of physiology fuction in organism, these important reason were resulted possibly in dead of minority chicks.But stage of recovery in organism could induced seceretory IgS of high level. The observations showed that humoral immune function in the chicks infected with AIV was enhanced. Thereby,ability of defense aggressive poison of the chicks were inhanced.The numbers of CD4+T, CD8+ T lymphocyte subpopulation, TANAE+ cells and IgA, IgG, IgM antibody-producing cells in immune issue of chicks infected with AIV were decreased compared with controls chicks 3~14 days PI in different degree. The results indicated that cellular immune function of chicks infected with AIV was decreased. The effects and mechanism of chicks infected with AIV were illuminated by the changes of Igs content, the number of Igs﹢producing cells and T cells in the experiment, which provides scientific guidance for defense of AI, which has significance in theory and practice.
