The present study is aimed at acquiring for the network by defining the effects of different concentrations follicular-stimulating hormone(FSH),androgen(An), estradiol (E2) and progesterone(P) on apoptosis, proliferation and steroidogenesis of granulosa cells(GCs) cultured in vitro, obtained from preovulatory follicles of buffalo, using flow cytometrey to investigate apoptotic incidence and proliferation index of GCs, and radioimmunoassay to evaluate the concentrations of progesterone and estrogen in culture media, dying with Hochest33342 to detect apoptosis of GCs cultured in serum-free media. The results showed that it was confirmed that GCs being cultured in serum-free media after 24 h, occurred apoptosis, which was dyed with Hochest33342. And apoptotic GCs exhibited bright blue on fluorescence microscope. Apoptotic ratio of GCs cultured in serum-free media on fluorescence microscope was consistent with flow cytometry.
