| Polygonum cuspidatum is abundant in resveratrol and piceid. The inducing conditions of Polygonum cuspidatum callus and the piceid accumulation during the cell growth as well as HPLC analysis method of resveratrol and piceid were studied using wild Polygonum cuspidatum in JinZhai of Anhui Province as experimental material.1. HPLC analysis method of resveratrol and piceid in Polygonum cuspidatum callus was established.2. The effects of hormone KT and 2,4-D on inducing callus were studied based on MS culture medium as basic condition. The result indicated that MS culture medium plus 0.8mg/L of 2,4-D and 0.2mg/L of KT was suitable for the inducement of Polygonum cuspidatum callus.3. The factors to affect the cell growth and piceid acuumulation during the cell culture were investigated. The resulted showed that pH 5~5.5, 800mg/L of Gln concentration and 4% of sucrose in culture medium was fit to cell growth and piceid accumulation in Polygonum cuspidatum callus. The fresh weight of callus increased 21.05 times within one growing period.4. The cell suspension culture of Polygonum cuspidatum was developed and the relationship between the piceid accumulation and cell growth was studied. The results showed that the piceid accumulation was not related with cell growth of Polygonum cuspidatum. The fresh weight and dry weight of Polygonum cuspidatum cell was 26.43 times and 25.48 times, respectively, compared to amount of inoculum cell within one growing period.
