Being a tropical fruit,banana(Musa spp.)is a typical climacteric fruit and has poor storage characteristics.Many researches have been focused on banana postharvest, storage physiology and molecular biology.So isolating and analysis of gene function in the fruit of banana's ripening are both a fundamental biological issue and a highly promising application.MuMADS1 has been cloned and identified as a group of important transcription activators of plants in our lab. The MuMADS1 gene was specifically expressed in banana flower and fruit, which was found to be associated with fruit ripening.In order to further study the MuMADS1, two plant expression vectors pBBBs were constructed in sense (pBBB-MADS1) and antisense (pBBB-aMADS1) orientation,and then were introduced into tomato by Agrobacterium tumefaciens-mediated transformation respectively.The transformed tomato might provide new base of material for the research of plant flower development regulation and fruit forming and ripening.A 1334 bp cDNA fragment was gained from the cDNA library of 2 days harvested banana fruit by using PCR technology, named MuBTB1.Sequence analysis and bioinformatics study showed that this cDNA encoded 109 amino acids, with a 807bp open reading frame(ORF) from 83 to 889bp.The result of BLASTx showed that MuBTB1 had highly homologous with other species BTB genes.The MuBTB1 was expressed in banana flower, leaf, root and fruit, highly expressed in flower and fruit by RT-PCR. The result indicated that the MuBTB1 was related with flower forming and fruit ripening.In recent years,small G proteins have become an intensively studied group of regulatory GTP hydrolyses involved in cell signaling, which take part in numerous and diverse cellular processes, such as gene expression, cytoskeletal reorganization ,microtubule organization, and vesicular and nuclear transport, functioning as molecular switches.Two primers were designed according to the positive recombinants obtained by yeast one-hybrid technique.
