Taian is the principal cow-producing area in Shandong province.Dairy industry is the mainstay of local economic development, which isalso an integrated agricultural system in the present situation. However,the annual average output of milk is about 4000-4500 kg. Except for the cause of the cow varieties, the mastitis is the main factor.In this study, We had a detection of subclinical mastitis on the 192 shares of milk which came from the suburb of huhhot by CMT. The results showed that 171shares were positive, the positive rate reached 46.4%. 171 strains of bacteria which include 6 kinds of bacteria were isolated from 171 shares, there were 86 strains of Staphylococcus(50.29%),37 strain of Streptococci (21.64%) and 29 strains of bacilli (16.96%).Multiplex polymerase chain reaction assay was developed for detection of Staphylococcus aureus and Streptococcus agalactiae causes of bovine mastitis. The PCR was optimized and the specificities and sensitivities of the PCR were analyzed. DNA extracted by boiling, 12 strains of Sta. aureus and 10 strains of Str. agalactiae were detected by the PCR. Expecting fragments were amplified from all strains. 49 milk samples from cows suffering subclinical mastitis were detected by the PCR and conventional bacterial identification. Staphylococcus aureus and Sh: agalactiae were detected by the PCR from 12 milk samples and 14milk samples, respectively.
