With the development and widespread application of biological technology,transgenic animals technology continues to mature and widely used in medicine, production, food, etc. It brings huge economic benefits to the society. Chinese has also won the breakthrough progress and gain a lot of scientific fruits. Transgenic rabbit, chickens, pigs, cows and other animals have come out and they have fast growth, strong resistance and good fleshy. However, transgenic animals likely have a negative impact on environment and biodiversity. The security question of transgenic animals has already become the focus of the society and the biotech security has become increasingly fierce, and the controversy is mainly concentrated in ecological environment, food safety, etc.In order to verify whether transgenic pig have influence to the environment, especially the exogenous gene whether drift to other organisms'genome. The paper carrys on safety assessment technology research of exogenous gene and transgenic pigs, focus on the key problems of safety assessment research. Aimed at the impact of transgenic pigs to environmental, we detect the gene drift between transgenic pigs and no-transgenic pigs, transgenic pigs and surrounding environment. We try to explore whether exogenous gene of transgenic pigs would be transferred to other organisms genome.1) Analysis on PCR detection ability of exogenous geneDiluted the known concentration plasmid which have been integrated exogenous gene and then amplified exogenous gene by PCR . The resμLts showed that the detection ability of appA,CMV-appA,MX1,CMV-MX1 in PCR system were: 2.4×103copies/μL,2.4×104copies/μL,2.4×102copies/μL,2.4×103copies/μL。2) Detection of gene drift between transgenic pigs and non- transgenic pigsExtracting DNA from placenta and blood of recipient pig and testing exogenous gene by PCR. The resμLts showed that there is no exogenous gene. Choose four transgenic sows and four Large White pigs in its 2 months and kept in a corral.Then collected the blood of four Large White pigs after 3,5,7 months and tested exogenous gene by PCR. The resμLts showed that there is no exogenous gene.3) Detection of exogenous gene in pig farm and soil around the pig farmExtracting DNA from feces in pig farm and confirming the inhibitory reaction of feces DNA to PCR.We amplified exogenous gene in 50μL amplification system with a certain amount of feces DNA to replace ddH2O.When we add 0,1μL,2.5μL,5μL,10μL,15μL, PCR detection ability were 2.49×105 copies /μL,2.49×106 copies /μL,2.49×107 copies /μL,2.49×108 copies /μL,0.We Extracted DNA from feces of transgenic pig,soil in pig farm and soil around the pig farm and tested exogenous gene by PCR. The resμLts showed that there is no exogenous gene. |
