Study On Tissue Culture And Rapid Propagation Of Yucca Gloriosa 'Variegata'

Yucca gloriosa 'Variegata', which belongs Liliaceae, Yucca genus, has very high ornamental value for its striking sword-like foliage edged with yellow and large inflorescence. This study was used terminal buds and suck buds as explant, the author established the tissue culture rapid propagation system, including the optimum disinfection method, sampling time, explants, the composion of primary medium, subculture medium and rooting culture, and the optimum training and transplanting method for the plantlets. The main results are as follows:1. The optimal explant surface disinfection method:Explant was disinfected with 70% ethanol for 15s, then treated with 0.1%HgCl2 for 10 min and washing by sterile water for 3～5 times, contamination is low, the germinateing rate is highest.2. The optimum sampling period:The explant was sampling in April, and the germinateing rate is high, contamination is lowest, and the sterile seedling grows quickly.3. Explant choice:Terminal buds and suck buds are best explant, and the germinateing rate is high, contamination is lowest, and the seedling grows well after the transplanting.4. Primary culture:Inoculated on primary medium which is MS+1.5mg/L+BA 0.1mg/L, the callus induction rate is 98%.5. Successive transfer culture:The best proliferation cultivate medium of Yucca gloriosa 'Variegata'is MS+6-BA3.0mg/L+NAA0.1 mg/L.6. Rooting culture:Roots were induced on the medium of 1/2 MS+IBA 0.5 mg/L, the rooting rate was 98%, the quantity of roots is 4.6, and the length is 3.6cm.7. Transplant and seeding:The more suitable acclimatization time is 10 days, and the survival rate after 30 days is 95%, the rooted plantlets were trained and transferred on the medium containing vermiculite, perlite and sand in the ratio of 1:1:2, which the rate of survival was 90%, and plants grow well.
