As an important cultivation technique, the Dwarfing culture and close planting can improve the yield and quality of Castanea mollissima, so they were attentioned by growers more and more, but there were no suitable cultivars. So rapid selection for new varieties for dwarf culture and close planting became the new breeding objectives. As a treasure germplasm, the weeping chestnut can let chestnut grow shorter. Since the conventional breeding cycle was long, in order to shorten breeding time, and speed up the breeding process, to lay the foundation for marker-assisted breeding and provide a way in the future,.Identification of molecular marker linked to the weeping branch traits of Chinese chestnut and Molecular Assisted Breeding was studied. The main conclusions are as follows:(1) In this research molecular markers linked to weeping branch trait were detected with AFLP and SRAP technique coupled with BSA method. in weeping branch varieties and vertical branch varieties, and 5 SRAP markers:me4Me1-160,me4Me1-120,me4Me2-240,me4Me2-130,me5Me2-200, which might be linked to the weeping branch traits of Chinese chestnut were screened.(2)According to the own structural characteristics of Chestnut seeds, the supporting nursering technology about the Marker Assisted Selection of Castanea mollissima Blume was determined, that is cutting 1/3 of seed for DNA extraction and sowed the remaining part.(3) Using the backcross hybrids, with the supporting nursering technology about the Marker Assisted Selection,2 SRAP markers:me4Me1-160,me4Me1-120, which might be linked to the weeping branch traits of Castanea mollissima Blume were definitized. And the validation results also showed that weeping branch traits of Castanea mollissima might be controlled by independent recessive genes.
