Porcine reproduetive and respiratory syndrome(PRRS) causes reproductive failure and respiratory diseases. In 2006, a HP-PRRSV variant which occurred in some provinces in south of China caused death of a lot of pigs. It spresd widely in china and made huge economic losses in pig industry of China. Reasons for the significant pathogenicity of HP-PRRSV are still unclear. In vivo, there will be differential gene expressions during HP-PRRSV infection. By using proteomics and bioinformatics we are likely to find important biomarkers and important protain.In the present study, we firstly compared test group infected with HP-PRRSV(HuN4) with the negative control group by using two-dimensional fluore-scence difference gel electrophoresis (2D-DIGE) and MALDL-TOF-MS/MS. And integrity proteomic data of mandibularis lymph node was obtained. A total of 24 proteins were significant alter between different groups by using 2D-DIGE, 22 differentially expressed proteins(DEPs) were identified by mass spectrometry identification. These proteins could be classified into the functions associated with morphogenesis, stress response, protein synthesis, Transcription, regulation. Of the altered proteins, four proteins, heat shock gp96 protein(HSPgp96), heat shock 70kDa protein (HSP70), heat shock 60kDa protein (HSP60)and MX1 involved in stress response or proteasome pathway were observed to be up-regulated(P≤0.05). Three morphogenesis proteins, Chain A The Crystal Structure Of Diferric Porcine Serum Transferrin, Chain B Crystal Structure Determination Of Porcine and Chain C Crystal Structure Determination Of Porcine were up-regulated. Three synthetic proteins: peptidylprolyl isomerase B was up-regulated, Chain A Structure Of Porcine Class Pi Glutathione S-Transferase was down-regulated, S-formylglutathione hydrolase was firstly up-regulated and then down-regulated. Three regulation proteins: 60S acidic ribosomal protein P0 was observed to be up-regulated, heterogeneous nuclear ribonucleoprotein A2/B1 protein(hnRNP A2/B1) was down-regulated, heterogeneous nuclear ribonucleoprotein H3(hnRNP H3) was firstly down-regulated and then up-regulated. Then analysis of expression of HSP70 in samples by Western bloting.This study aimes to establish HP-PRRSV infested lymph nodes protein map, finding differences protein expression for new diagnostic markers signaling molecules. The results we gained might be a valuable clue for further HP-PRRSV pathogenesis mechanism study and research.
