| Resurrection plants are a group of species that can tolerate extreme dehydration and survive when water becomes avaible. Boea hygrometrica is widely distributed in southwest and north China. It can lose more than 95% water when the weather is drought, but it can resurrect when water is avaible. In this paper, the changes of electrical conductivity,photosynthetic activity and ABA content in the process of drought were determined. The results showed that electrical conductivity was increased, photosynthetic activity and ABA content was declined during dehydration, but these parameters resumed after rehydration.Creeping bentgrass is a major turf-grass species in North China. To provide an efficient platform for turf-grass breeding using transgenic approach, the condition to induce callus from creeping bentgrass seeds were optimized. The results showed that the callus inducement could be increased to 90.72% when the seeds were treated with 50mg·L-1 GA3 for 4ds at 4℃before germination and the callus induction media was MS+2,4-D 5mg·L-1+ casein hydrolysate 0.25g·L-1+praline 0.25g·L-1.BhGolS1, a drought inducibe gene encoding galactionol synthase (BhGolS1) from B. hygrometrica, had been shown to improve the drought tolerance of transgenic tobacco efficiently when overexpressed. In order to improve the drought resistance of turf-grass creeping bentgrass, a monocot expression vector p3300-ubi-BhGolS1-Tnos was constructed and transformed into creeping bentgrass callus via the method of Agrobacterium-mediated transformation. By antibiotics selection with Basta spraying and PCR identification with gene specific primers, it was verified that the gene was successfully transformed into creeping bentgrass and the positive plantlets were obtained. |
