This study belongs to the Qinghai-Tibet Plateau in Shiqu County, Luhuo county, Dege county, Daocheng, Ruoergai County, Hongyuan County Sichuan provincial and Zeku County,Gangca County, Qinghai Province, Gansu Province, Qilian County, Luqu County and Tibet Lhasa, Nagqu County, Yangbajing, Damxung County and other 14 regions of the wild mushroom resources on the line in the survey to understand the Tibettan Plateau genetic diversity of wild Agaricus bisporus. Advantage of the collected strains of wild mushrooms combined with the genetic differences between cultivars were analyzed with the mycelial growth rate at the same time, temperature measurement experiment as well as some strains carried out in different parts of fruiting tests to extensive breeding of Agaricus bisporus. Individual collected in different strains of rDNA 18s region were amplified withITS4-ITS5 and compared with the known species of mushrooms Blast sequence in the GenBank to get a higher ITS sequence homology, and mushrooms of the genus Other types of phylogenetic tree was constructed as a reference. The results showed that: AGQL811 sequenced strains and Agaricus campestris clustered together of the 99% confidence level, the other 7 sequenced strains and Agaricus bisporus were clustered together of the 99% confidence level.Inter-simple sequence repeat amplified polymorphism (ISSR) technique, using 9 primers on the Tibetan Plateau wild mushrooms strains 79 and 10 control strains were genetic diversity of strains constructed by genetic correlation clustering Fig. The results showed that: the genetic similarity coefficient of variation range is 0.24-1, when the similarity coefficient keeps 0.46, the 89 isolates clustered into three groups. The first 83 strains, 10 control strains were gathered in the first class, second class has four strains, there are two strains in the third category. 79 wild strains and 10 control strains did not appear in exactly the same genetic strain background, indicating that the wild strain in the DNA has a relatively significant level of genetic variation, showing that the rich genetic diversity. ISSR analysis and morphological characteristics combined with identification of the first type of wild strains is Agaricus bisporus, the second category of wild strains is Agaricus campestris.Some strains in different regions of the mycelium growth rate were studied, the results showed that the growth rate of wild strain is slower than the growth of control strains.Some strains of the different regions were carried out with fruiting test, the results showed that the strains of wild mushrooms emerged brown color, yield lower than the control cultivars, and significant differences.
