| Objective:Ginseng Angiotensin I-converting Enzyme Inhibitory protein which have powerful biological activities but less reasearches,were studied by technologies including isolation and purification technology,activity detection technology and analysis-identification technology. Through studied on extraction, purification and activity of Ginseng Angiotensin I-converting Enzyme Inhibitory protein,providing a basis for studies on ACE inhibitors.Methods:Using different ratio of solution,different extraction time and different extraction times extract the ginseng protein and ginsenoside,using Coomassie brilliant blue G-250 method and Acetic Acid-Perchloric acid method,determin the rate of protein and ginsenoside extraction for the evaluation Standards,choose the best optimized extraction process for Orthogonal test.Purification was carried by column chromatography technology,combined with Inhibitory activity to screening activity component by UV spectrophotometer method.Compare the difference of the proteins among ginseng protion and Ginseng Angiotensin I-converting Enzyme Inhibitory protein by using Tricine-SDS-polyacrylamide gel electrophoresis(Tricine-SDS-PAGE).Results:The single factor experimental results show that different ratio of solution,different extraction time and different extraction times were all have an affect to the extraction rate of the total protein and total ginsenoside. Orthogonal experimental results,the highest is liquid ratio of 1:15,extraction time of 12h and extraction times of 3 times.Have the higest yield of protein,yield up to 3.41%.And have higer yield of ginsenoside,yield up to 3%.Ginseng protein extract was hydrolyzied by NaOH (1mol/L), harvestied the Ginseng Angiotensin I-converting Enzyme Inhibitory protein. ACE inhibitory activity experiments showed that it have higer inhibitory effect, inhibitory ratio up to 92.5%.And electrophoresis results showed that the main protein of ginseng were distribution in the 60-70KDa and 30-35KDa.however,the Ginseng Angiotensin I-converting Enzyme Inhibitory protein were distribution in the 10KDa.Conclusion:The best extraction process of ginseng total protein is:Distilled water as solvent,liquid ratio of 1:15,extraction time of 12h,extraction times of 3 times and extraction temperature of 4℃.It solvedroblems of complicated operation,long cycle and higher costs during traditional process and it is suitable for industrial production.The use of column chromatography technology will make a high inhibitory activity of the Ginseng Angiotensin I-converting Enzyme Inhibitory protein, and molecular weight distribution in the lOKDa. Eventually build a quick, simple and efficient purification process route for further research of the mechanism of ACE aspects of ginseng research and new drug development to lay the foundation. |
