In this thesis, Extracted polysaccharides from Shaggy Mane by boiling water teaching and ultrasonic-assisted process, then extracted polysaccharides were isolated, obtained Shaggy Mane polysaccharide API and Shaggy Mane polysaccharide CAP1. Shaggy Mane polysaccharide API was structural analysised. Then analyse and comparison of Polysaccharides API with CAP1. The affect of ultrasonic-assisted extraction on the structure of Shaggy Mane polysaccharide AP1 was investigated. Main contents and results as follows:(1) Extracted polysaccharides from Shaggy Mane by boiling water teaching and ultrasonic-assisted process, then extracted polysaccharides were isolated and purified through the precipitation with cetyltrim-ethlammonium bromide (CTAB),Sephadex G-150 gel filtration. Used high performance liquid chromatography (HPLC) to identify the purity of Shaggy Manepolysaccharide AP1 and Shaggy Mane polysaccharide CAP1, proving that they both were homogeneous polysaccharides. High performance gel permeation chromatography determined the molecular weight of Shaggy Manepolysaccharide API was 5.4×105 Da,the molecular weight of Shaggy Mane polysaccharide CAP1 was 4.1×105 Da.(2) Analyse Polysaccharides API extracted from Shaggy Mane by boiling water process.By means of HPLC,GC,FTIR,GC-MS,partical hydnolys is with acid,periodate oxidation,Smith degradation,methylation analysis and Atomic force microscopy, the structure of API was identified.The results indicated that API was composed of mannose, glucose, galactose. Infrared spectra showed that polysaccharides API had saccharide characteristic absorption peaks in the range from 4 000 to 650 cm-1.1→3, 1→6glucose residues in main chains,1→3,6 glucose,1→4,1→6 galactose and 1→4,6 mannose in side chains. AFM image of Shaggy Mane polysaccharide API showed AP1 Has the branching structure.(3) Analyse and comparison of Polysaccharides API with CAP1 extracted from Shaggy Mane by boiling water teaching and ultrasonic-assisted process were carried out.The affect of ultrasonic-assisted extraction on the structure of Shaggy Mane polysaccharide API was investigated. The results showed that the components of Polysaccharides AP1 is identified by GC as:mannose, glucose, galactose; and CAP1 is xylose, mannose, glucose, galactose; Infrared spectra showed that polysaccharides API and CAP1 had saccharide characteristic absorption peaks in the range from 4000 to 650 cm-1.The infrared spectrum of API had absorption peaks at 865 cm-1 and 764 cm-1,but CAP1 had the peaks at 903 cm-1,826 cm-1 and 756 cm-1.Atomic force microscopy diagram showed the polysaccharides had branch structure; The influence of ultrasound was likely to cause the connection pattern change among monosaccharides,and easily resulting in the formation of relatively uniform mesh structure.
