In the experiment,in order to analyse the methods of identification of resistance to Verticilium wilt in cotton in greenhourse,31 cotton cultivals were used to compare the speed,period and veracity of the methods of inoculation.Tissue structure was studied to realize the resistance mechanism of cotton to Verticilium wilt.The RGA primers were used to find out the primer closely linked with resistance gene,which could be as function primers to serve for the identification.The main results were as follows:SHZ-134 separated from 134 corps in Shi Hezi City was compared with the strains which pathogenicity were known.The result showed that V76 and BA were defoliating strains.SHZ-134 and TS-2 were non-defoliating strains,having medium pathogenicity.The pathogenicity of 3H were the weakest.Among the four inoculative methods, the speed of the leaf pricking and spreading was the fastest.The speed of the root-dipping was the second,and that of irrigation was the slowest.Compared with the field appraisal, the result of root-dipping was the most consistent with that by cut-stalk in field, wheras the result of irrigation was the worst.There had not significant difference between the result of root-dipping and cut-stalk.The root-dipping was the best, through estimating the time, period and the result.The density of roots that absorbed water and rich had correlation with the disease index, r=0.923.There was significant difference among the resistance,the tolerance and the suscept.But the difference between the tolerance and the suscept were not significant.The resistance had the biggest density of xylem parenchyma cells ,the most vesselses and the smallest diameter in stem and root.But all the items were contrary in the suscept.Using 107 pairs of the RGA primers to amplify in parents and the F3 group, the result showed that three primers were the most consistent with the result by cut-stalk, and it could be concluded that the three primers were closely linked with the resisance toVerticillium wilt.
