The microsatellite markers were used in this experiment to analyze the genetic diversity of six Tibetan sheep breeds in Gansu and Qinghai Provinces (Minxian Black Fur sheep, Oula Tibetan sheep, Qiaoke Tibetan sheep, Ganjia Tibetan sheep, Guide Black Fur sheep, Qinghai Plateau Tibetan sheep) and one control breed (Tan sheep) at eight microsatellite loci. The objectives were to offer the scientific data for protecting and utilizing sheep genetic resources. Main results were as following:1. By determining genetic diversity of 8 microsatellite loci in 7 sheep breeds,103 alleles were detected, the average alleles at each loci were 12.9. There were the most alleles at the locus OarFCB128, while the least alleles at the locus MAF70. So, Tibetan sheep breeds in Gansu Province had rich genetic diversity.2. As for 8 microsatellite loci in 7 sheep breeds, the polymorphism information content ranged from 0.851 to 0.878, the heterozygosity ranged from0.876 to 0.898. Therefore, in this study, the 8 microsatellite loci analyzed were highly polymorphic, which can be used to evaluate the genetic diversity in sheep.3. Nei's genetic distance ranged from 0.1103 to 0.2427, Nei's standard genetic distance ranged from 0.1375 to 0.3232. The genetic distance between Minxian Black Fur sheep and Guide Black Fur sheep was the smallest(DA=0.1103,Ds=0.1458).The genetic distance between Qinghai Plateau Tibetan sheep and Tan sheep was largest(DA=0.2427,Ds=0.3056).4. 7 sheep breeds were clustered into three groups. Qinghai Plateau Tibetan sheep, Qiaoke Tibetan sheep, Oula Tibetan sheep, Ganjia Tibetan sheep were grouped. Minxian Black Fur sheep, Guide Black Fur sheep were grouped. Tan sheep.
