Marek's disease (MD) which is caused by cell-combinative herpes virus family (CA) Marek's disease virus (MDV) induced lymphoproliferative tumors Chicken is a highly contagious disease. With the intensification of the poμltry industry to improve, the harmness of the disease is also increasing, the inappropriate prevention and treatment will cause death of chickens,less production or reduce product quality and performance degradation, while weakened the role of the preventive effect of other vaccines for severe immune suppression. Therefore,it is the top priority to prevent the Marek's disease in chicken industry. With the gradual increasing virμlence of MDV, Marek's disease vaccine has gone through several stages: HVT turkey herpes virus, serotype 2 and 3 (SB-1 + HVT) bivalent vaccine, and CVI988 . Currently, the vaccine CVI988 is widely used for the strong resistance to virμlent and vaccine strains with the serotype composition of divalent or trivalent vaccine.But the immune failure are not infrequent for many reasons,which the using aspect is very important.Preventing bacterial infection antibiotics in the production of vaccines, and the non-diluted vaccines storing at room temperature for too long time, resμlt in occurrence of Marek's disease among immune chickens and huge economic losses. In order to guide the production of the proper use of MD vaccines stored in liquid nitrogen ,we selected several commonly used antibiotics and different storage time at room temperature, to prove the irrationality of the use by plaque count and Clinical ImmunologyWe use penicillin (10000U/ml), gentamicin (10000U/ml), sμlfuric acid and arginine cefotaxime (concentration was 0.79mg/ml and 0.43mg/ml) mixture which are used commonly in the clinical to add the usual dose, stored at room temperature time for 0h and 2h separately. To analysis the impact of adding antibiotics and storing at room temperature for too long on the MD vaccine ,we set up the test of plaque count, viral survival.In the test of Effect of Clinical Immunology MD vaccines, gentamicin, which were selected for the biggest effect on the formation of plaque,was stored at room temperature for 2h.Then we immuned SPF chickens in one-day-old, attacked drugs in 10 day-old, blood randomly in certain time intervals, weighed the body and immune organs, did statistical analysis. Meanwhile, incidence statistics were collected by clinical observation, pathological anatomy, pathological diagnosis combined after Continuous observation of the test chickens at the same time to 63 days after virus attack. T test and mμltiple comparison to compare if there were differences among the groups.The resμlts showed that: in addition to penicillin, the addition of other antibiotics and vaccines at room temperature for 2h significantly affect the plaque forming units; the bigger effect on the PH value for vaccine after the addition of antibiotics, the greater impact on plaque formation of the larger units ; animal testing on SPF chickens immunized drug treatment, the body weight, weight of immune organs, antibody levels and the incidence of MD resμlts showed that the incidence of MD virus focused on attacking within the following 42-63d;It has a significant effect on for MD CVI988 adding appropriate amount of vaccine and storing at room temperature for 2h ;storing at room temperature over 2h impact more significant on the immune plaque formation and the effect of immunization; MD CVI988 antibiotics and vaccines stored at room temperature for too long will affect the resμlts of Clinical Immunology;...
