Inhibition Effect Of Curcuma Oil Injection On Porcine Parvovirus In Vitro

Curcuma oil was extracted by Supercritical CO2 method (SFE method), the optimized condition is the granularity 60, the temperature 55℃, and the pressure 25Mpa. The oil was extracted from Curcuma. Curcuma oil composition with GC-MS analysis had two kinds of unknown composition except curcumol, Curcuma ketone element and other components. The Curcuma oil was deluting to 1% with 2% Tween-80.Curcuma oil effect on PK-15 cells, TC0 was 62.5μg/mL by the CPE method, and TC50 was 125μg/mL; TC0 of the MTT was 28.67μg/mL and TC50 was 49.33μg/mL. The effect of prevention, treatment and disinfection of Curcuma oil of TC0 concentration with MTT method was used to research its mechanism of anti-virus. Firstly, PK-15 were added drugs , then added the virus, detecting whether drug could combine with cell surface receptors and prevent virus to adsorpt and entry; Secondly, PK-15 was added virus, then added the drugs, observing the effect of the drugs inhibition virus in cells; The end, virus and drugs was added together, observing the direct inactivation. It showed that the best way to inhibit the virus is the first one among three methods. Since IC50 was 2.813μg/mL, IC90 was 4.123μg/mL, and therapeutic index was 31. Compared with the positive control, it is significantly (P<0.01), but there was no difference with negetive control.Use to add Curcuma oil 25ug/ml and then add PPV 100TCID50, collect the infected-cells and culture in different time, then detect the PPV copy volume by real time quantitative PCR. Compared with the positive control, discussed the proliferation rules of Curcuma oil inhibition PPV copy in the effective concentration. The results of ANOVA showed that compared with the virus control (P<0.01). DNA copy of the virus was not significant (P>0.05) after 2-12h, 12h later PPV copies of viral DNA (P<0.05), 65h later PPV DNA proliferation (P<0.01).Curcuma oil could inhibite porcine parvovirus.The 4.123-28.67μg/mL of curcuma oil could be used to therapy and privent, when the PPV vaccine wouldn't make immunoprotection and PPV .
