The three populations of coilia ectenes on the Yangtze River in Anhui Province were studied by using morphology and molecular methods.The morphology of coilia ectenes were studied through comparisoning of variable means,length and weight compositionOne—Way ANOVA and cluster analysis. The results showed that there were similarity between the three populations of coilia ectenes,Wuwei population was close to Anqing population,but it was discovered crossing between Individual variable.From the relation between length and weight of coilia ectenes,there was no significant boundary between stocks,but swarm of individuals.The mitochondrial DNA COI and Cyt b gene fragments of Wuwei,Anqing,Dangtu populations were amplified and sequenced(The number of samples of COI and Cyt b gene were 58,57 respectively),then analyzed and compared the differences between groups and individuals. The sequence length of the COI gene partial sequence is 606bp,and there was three deletion sites,nine variation sites,and uneven distribution on populations. Nucleotide composition analysis revealed that cytosine content was lowest(17.97%),The G+C content was lower than that of A+T. The genetic distance(Pairwise distances)of individuals was 0.0000-0.0165.Then the UPGMA and NJ phylogenetic tree was constructed based on Kimura-2 pattern, and revealed that crossing was existed between Individuals. Search and download COI gene partial sequence of coilia nasus,Engraulis japonicus,Salamandra salamandra,Mus musculus and Homo sapiens .Then the UPGMA and NJ phylogenetic tree was constructed based on Kimura-2 pattern of above five species and three populations of coilia ectenes. And the phylogenetic tree revealed that Wuwei population was close to Anqing population,which are consistent with the result of morphological analysis. The sequence length of the Cyt b gene partial sequence is 410bp, no insertion or deletion of site was found, just 4 variation sites,and all variation sites changed thymine(T) for cytosine(C). Nucleotide composition analysis revealed that guanine(G) content was lowest(15.66%),The G+C content was lower than that of A+T. The genetic distance(Pairwise distances)of individuals was 0.0000-0.0074.Then the UPGMA and NJ phylogenetic tree was constructed based on Kimura-2 pattern, and revealed that parallel relationship was existed between Individuals, difference was obvious of individuals between populations. Search and download Cyt b gene partial sequence of coilia nasus,Engraulis japonicus,Salamandra salamandra,Mus musculus and Homo sapiens .Then the UPGMA and NJ phylogenetic tree was constructed based on Kimura-2 pattern of above five species and three populations of coilia ectenes. And both of the phylogenetic tree revealed that Anqing population was close to Dangtu population.Through the study on morphology and molecular of coilia ectenes, it revealed that relationship between three populations of coilia ectenes:Wuwei,Anqing,Dangtu were very close. There are no significant differences among the three groups on morphological characters except the Variance of D(HL/SL)and H(BL / SL);The phylogenetic tree showed obvious crossing between individuals. Preliminary study on coilia ectenes in this paper is order to offer theoretical basis for rational exploitation and utilization of coilia ectenes germplasm resources ,and fill up the blank in the field of coilia ectenes germplasm resources of Anhui province,which is important to protect the genetics diversity of coilia ectenes.
