The corn is one of the most main three grain crops in the world, simultaneously it's also playing its vital role as feed. Since the 21st century, the method of using the plant genetic engineering's to obtaine the new corn variety has been great concern. But the plant genetic engineering method has had many problems, such as the high cost , the complex operation, low conversion rate and so on.The paper aims to obtain transgenic maize by genetic engineering technique, which makes the anti-weed killer gene bar transfer to the maize by pollen-tube pathway. At the same time it analysises and improves the transgene's sequence of operation and raise the conversion rate. As there is still much controversy about pollen-tube pathway, this paper bases on the theory which the agricultural bacillus can infect to the maize to transgene to propose new methods: (1) the transgenic methods of injecting in the coleoptiles (2) the transgenic methods of cutting coleoptiles (3) the transgenic methods of cutting seedling. After the instantaneous expression and examination of the mark gene gus, it explored and studied the feasibility and the technical question of this method. When the length of the maize plumule is 2-3mm, the most suitable vaccine density of agricultural bacillus namely OD is 0.6-0.7, the days of co-culture is 3, the gus gene expression conversion rate by the jet method is the highest. These provide the new corn conversion system's establishment with the reference.This article further conductes the research to three transformed methods, observes the deformity of the stem and leaf during the transformation process. through the initial screening by herbicide of resistance bar gene the transgenic maize plants appear. Clear that only the injury appears in the growing point cells , the extent of damage is two acupuncture, the survival rate and conversion rate is high. And the conversion rate is 9%. Maize materials which have been transformed for 3 days are observed by paraffin sections method.Prove the feasibility of three methods from the morphogenetic point of view. The study of transgenic maize provides new ideas and methods to streamline operations, improve the conversion rate. And it also has important theoretical and practical significance.
