Atractylodes lancea (Thunb.) DC. is a kind of herbaceous perennial in Atractylodes of Asteraceae family. Its rhizomes are used as the traditional Chinese medicine, which have many effects as drying dampness, strengthening the spleen, relieving rheumatic colds, dispersing cold, improving eyesight and so on. The wild A. lancea reproducted by seeds, but because of lower seeds repeningrate, lower seedling rate and longer production cycles, the cultivated is mostly depended on asexual reproduction, but which used for many years always cause serious degradation of quality such as yielding ability, resistance ability, quality and so on. Production can not meet the demand of planting expansion. It is necessary to study on the breeding technology of A. lancea seedlings. It will not only improve the survival rate of A. lancea seedlings, cut down planting costs and increase revenue, but also is conducive to rejuvenation and breeding good cultvars. This paper studys on the herb seeds (seedlings) standardization, key cultivation techniques of seedlings and storage technology of 1-year seedlings and other aspects. The results are showed as follows:1 By thousand-grain weigth,germination rate,water content and purity degree of local seeds of A.lancea, the grading standard of seeds is described as the 1st,2nd and 3rd grades. Purity degree of any grade should not be less than 90%, water content 7.0%-7.5%, first grade, germnation rate≥90%, thousand-grain weight>13g; second grade, germnation rate80%-90%, thousand-grain weight 9g-13g; third grade, germnation rate< 80%, thousand-grain weight<9g.2 Studying the influence of germinating bed and temperature to seed germination, the results indicate that on the perlite bed the seed germinability of A.lancea reaches to 55.24%, which most significantly better than on sand bed, and germination rate is the best to 84.84%; the germinating rate in 22℃is best to 94.67%, most significantly better than which in 15℃or 5℃alternating to 15℃.3 By soil treatment, seed treatment, cover methods after the sowing, spraying EM(Effective Micro Organisims) during seeding stage, chemical control of the black spot disease and so on, a set of key seeds cultivation techniques were obtained. The fine soil mixed with 80% carbendazim WP being spread to seedbed by 12g/m2 can reduce soil pathogens and raise the seedlings emergence rate. The seeds germination potential in field and germinability indoor was significantly improved by soaking seeds 2h with 200 times EM. Covered with huofei and straw after sowing help to maintain soil moisture and temperature and increase the herb seedling emergence, seedlings rate, yield and weigth of single rhizomes. Spraying with EM during seeding stage can strengthen the seedlings, reduce the mortality of seedlings, and accelerate heigth grown; 500g/L Iprodione SC in 1000-fold can effectively prevent black spot disease about 80% control efficiency, reduce the mortality of seedlings and increase the yield, followed by 70% propineb WP in 500-fold which can effecticely prevent disease about 46%, increase seedlings rate, and can promote the growth of A. lancea rhizomes.4 By indagating the weight and the number of buds per seedlings, the rhizomes of 1-year seedlings of A.lancea is described as the 1st,2nd and 3rd grades. The first grade, weight per seedlings≥3.5g, buds≥5; second grade, weight per seedlings 2.5g-3.5g, buds three or four; third grade, weight per seedlings<2.5g, number of buds is one or two.5 The results of store experimentation of 1-year seedling'rhizomes indicate that the survial rate after 80d by storing in sand and hang outside is about 94% and 92%, by storing in sand after soaking rhizomes 15min by 80% carbendazim WP 1000-fold is about 97%.
