Er-Zhi-Wan (EZW), a famous traditional Chinese formulation, has been developed as arestorative formula for hundreds of years. It possesses the actions of tonifying the liver and kidney yin and widely used in China to treat kidney and Alzheimer’s diseases, invigorate the body, and modulate the immune system in clinical for many kinds of application.In this study, mechanism of EZW’s anti-osteoporotic was clarified. After treated with MSG, the mice appeared to a typical osteoporosis symptom, while after administrated with EZW, the EARE, BMC, and BMD data showed that osteoporosis symptom was allevited. Hypothalamus nerve cells and estradiol from osteoporosis mice were also tested after treated with EZW, and the results showed that the anti-osteoporotic activity depends on its restraining on apoptosis of hypothalamus nerve cell and improving on estradiol.The proliferations or differentiations of cells related to osteoporosis were determined after treated with EZW-containing serum. The proliferation of osteoblasts, cell lines UMR106, RAW264.7 as well as the differentiation of RAW264.7 to obsteoclasts was teated by MTT or TRAP assays. Results were demonstrated that its antiosteoporotic activity carried out by restraining osteoclastic bone resorption.Meanwhile, fingerprints were compared between EZW-containing serum and EZW extraction following serum chemistry method, results showed that its anti-osteoporotic activity is related to constituents whom belong to high polarty parts.Extract conditions for maximizing oleanolic acid and ursolic acid recovery were investigated. The concentration of EtOH, time, and volume of solvent were all considered, the results which were managed by uniform design, suggested a final optimizing extract method.Extract conditions for maximizing its antiosteoporotic activity were arranged via serum pharmacology method. Considering on the proliferation of RAW264.7 and differentiation of RAW264.7 to obsteoclasts, extract solvent (0%-100%EtOH) were compared in vitro, data showed that extracts from 0%-40% EtOH performed good anti-osteoporotic activity.
