Objectives1. To establish the post-traumatic stress disorder SD rat model of "fetus deficit" on the theory of Traditional Chinese Medicine.2. To observe the effect of post-traumatic stress disorder "fetus deficif" environment on offspring’s macroscopic characterization, such as growth and behavior ability.3. To observe the effect of the rich environment combined with emotional care nursing on offspring.Methods36 SD rats were divided randomly into three groups, emotional group, PTSD group, blank group (n=12).The emotional group was given emotional nursing after post-traumatic stress disorder modeling; The PTSD group was given no nursing measures after modeling; The control group were not given post-traumatic stress disorder molding and other measures just like blank group.The pregnant rats’s weight and behavior were observed in the pregnancy stages; the offspring rats’s growth indexe and behavior ability were measured, such as body weight, body length, tail length, Eyes Opening(EO),Turning Over Test(TOT), Open Field Test(OFT), Elevated Plus Maze Test(EPM).And also the 5-HT and GH level in offspring rats’s blood were determinated by ELISA.Results1. Growth TestThere were no significant difference between emotional group and blank group at different time points on the offspring rat body weight, body length and tail length(P>0.05); There was a statistically significant difference between blank group and PTSD group, the blank group offspring rat body weigh, body length and tail length were better than that of PTSD group(P<0.05); The emotional group compared with PTSD group, the offspring rats of emotional group body weight were better the offspring rats of PTSD group, the difference was statistically significant(P<0.05); At day 10, day 20, day 30 in birth, the emotional group compared with PTSD group, the former offspring rat body length, tail length were better than the PTSD group, the difference was statistically significant(P<0.05);The TOT and EO results had statistical difference between blank group and PTSD group,emotional group and PTSD group(P<0.0167).2. Behavior TestThe TOT results had statistical difference between blank group and PTSD group,emotional group and PTSD group(P<0.0167); In the OFT, the number of climbing lattices had statistical difference between control group and PTSD group, emotional group and PTSD group(P<0.05); EPM test results showed that PTSD group and emotional group, PTSD group and control group, there were significant difference between OE%(P<0.05), emotional group and PTSD group, PTSD group and control group, there were significant difference between OT%(P<0.05).3. Biochemical TestOffspring rats 5-HT levels distribution show the following trend:emotional group> control group>PTSD group, and emotional group compared with PTSD group, there was a significant difference of the 5-HT level (P<0.05); emotional group compared with PTSD group, there also was a significant difference of the 5-HT level (P<0.05).GH levels distribution show the following trend:control group> emotional group>PTSD group, and the emotional group compared with PTSD group,there was a significant differences of the GH level(P<0.05); control group compared with PTSD group, there also was a significant difference of the serum GH level(P<0.05).Conclusion1. Post-traumatic stress disorder "fetus deficit" environment could prevent rat offspring growth and development, and the offsprings of behavior ability and biochemical levels produce;2. Emotional Nursing could have protective effects on the post-traumatic stress disorder induced rat offspring growth and development of retardation and the reduction of behavior ability.
