Micro-environment of cells is capable of regulating cells’ daily activities, like protein expression, mitosis, differentiation, apoptosis etc., through physical or chemical factors. Among physical factors, the topography of the micro-environment is attraction more and more attentions. In case of bone, there is a multi-scale network composed of collagen, proteoglycan complex and other organic components. This network plays an indispensable role in the process of maintaining physical and chemical properties of bone, and regulating the functions of various bone cells. Thesis dissertation focused on the topography of the organic phase of the bone excellular matrix, aiming to promote the understanding of the bone micro-environment through this particular perspective. Firstly, we characterized the structure of the network by AFM and SEM; Secondly, we established a method based on image analyzing algorithm to extract geometric parameters of the random fibrous network; Thirdly, we made a computer geometry model and a micro-scale electrostatic spinning fiber invitro model; Finally, we build a nano-scale invitro model made of hydrogel and studied the effect of tethering degree of the nano-scale network on the spreading shape of mesenchymal stem cell. This dissertation may provide a basis for further study of the role of the mutiscale network in the physiological and pathological process, and for designing more reasonable bioengineer material.
