Purpose:To investigate the effect of total glucosides of paeony on rabbit knee osteoarthritis modelhistomorphology and synovial matrix metalloproteinases influence.Material and method:40 rabbits were randomly divided into 4 groups, 10 rats in each group, which is labled as normal group, model group, control group(ZW) group and treatment group(TGP) group, respectively. By using the improved Hulth classical modeling methods for modeling. Give control group: the Zhuangguguanjie pill 1.0 g/kg physiological saline 10 ml/time, 3 times/day; the treatment group: with total glucosides of paeony 1.0 g/kg physiological saline 10 ml/time, 3 times/day of drug orally drug, after 4 weeks, observe the change of synovial joints in thenaked eye. The expression detection of synovial MMPs was tested by the immunohistochemical method.Results:1. The naked eye can see, the group of normal articular cartilage surface is smooth, pale bluecolor, with cartilage, no crack and softening change, and synovial tissue without hyperplasia and thick and sticky even changes. In model group, articular cartilage surface rough, yellow,loss of luster, surface roughness, subchondral bone edge there are small osteophytesformation, synovial exist different degree of hyperplasia, thickening and adhesionphenomenon, visible in the synovial hyperemia. The control group and the treatment groupalso showed small crack surface of articular cartilage and synovial hyperplasia, which controlsthe articular cartilage surface pale yellow. The treatment group cartilage color ispale, the most obvious effect. 2 Normal group: synovial lining cell layer only in a small number of MMP-1, 3, 9 positive cellsdistribution; model group: can lining and its lower mononuclear cells in cytoplasm, see MMP-13, 9 positive cells, granular or lump, as light brown yellow to brown; control group: lining celllayers visible MMP-1, 3, 9 positive cells, showed weakly positive in cytoplasm, as light brown yellow; treatment group: lining cell layer visible MMP-1, 3, 9 positive cells, showed weakly positive in cytoplasm, as light brown yellow. 3 By image analysis software. The results show, the model group MMP-1, 3, and 9 levels were higher than normal group increased significantly, the difference was statistically significant(P<0.01). Group and treatment group MMP-1, 3, 9 levels lower than the model groupcompared with statistical difference(P<0.01). The treatment group compared with control, can reduce the content of MMP-1, 3 and 9, there were significant differences(P<0.05).Conclusion:Total glucosides of paeony have significant control effect on knee osteoarthritis, clinical effect is better than that of Zhuanggu Guanjie pill.
