Objective:To observe the effects of different doses of Radix curcumae extract learning on change the P2X4R/BDNF L4-L6segments of the spinal cord of CCI rats. Discussion on P2X4R/BDNF pathway of Radix curcumae extract stress mechanism of CCI rat model of neuropathic pain based on.Methods:1. Male SD48rats (body weight250-294g), were randomly divided into4groups,12rats in each group:normal group (group N), CCI modeling for third days group (group C3), CCI modeling for seventh days group (group C7), CCI modeling for fourteenth days group (group C14); TWL and MWT were determinated before modeling and modeling3,7,14days; BDNF, IL-1β content were measured by the method of ELISA outer peripheral blood; Expression of P2X4R and P-P38were measured in L4-L6spinal cord of rats by Western blotting; BDNF content of in L4-L6spinal cord of rats were measured by Immunohisto-chemical method in each group.2. Male SD56rats (body weight250-294g), were randomly divided into normal group (group N)(n=8), CCI model group (n=48); to be successful model after CCI in model group were randomly divided into6groups,8rats in each group:CCI model group (C group), Radix curcumae extract low dose group (60mg/kg; group Y1); Radix curcumae extract middle dose group(120mg/kg; group Y2); Radix curcumae extract high dose group (180mg/kg; group Y3); gabapentin group (100mg/kg; group G); solvent group (group S);each group according to different drug orally once every day. Measurement of TWL and MWT were administered at the fourteenth days; the content of BDNF, IL-1β were measured by the method of ELISA outer peripheral blood; Expression of P2X4R and P-P38were measured in L4-L6spinal cord of rats by Western blotting.Results:1.1The CCI model of body mass in rats decreased significantly than the normal group (P<0.01);in the fourteenth day of modeling, rats in the normal control group body weight is greater than the model group (P<0.01). After the model rats of third days, seventh days, fourteenth days he mean value t of TWL and MWT decreased, and significantly less than normal rats (P<0.01), suggesting the model is successful.1.2The concentration BDNF, IL-1beta in the peripheral blood of model after3,7,14days rat were markedly elevated compared with normal control group (P<0.01); model after3,7,14days in L4-L6spinal cord of rats, the expression of P2X4R/P-P38in the spinal cord gray scale value and reference ratio were higher than normal control group increased significantly (P<0.01). BDNF immunostaining semiquantitative scoring product of the model after3,7,14days rat L4-L6spinal dorsal horn were markedly elevated compared with normal control group (P<0.01).2.1Administration for fourteenth days, Radix curcumae extract body weight of rats in high dose group is lower than that of rats in the normal control group (P<0.01), but higher than the other5groups of rats (P<0.05or P<0.01); compared with the solvent group, high dose of Radix curcumae extract group, gabapentin group could significantly prolong the TWL value (P<0.01, P<0.05) and MWT value (P<0.01, P<0.05). And the high dose of Radix curcumae extract group TWL had no significant difference between the normal group (P=0.808).2.2Administration for fourteenth days, compared with the solvent group, middle dose group, high dose group of Radix curcumae extract,Gabapentin group BDNF, DL-1β concentrations in serum were lower (P<0.01;or P<0.05); and the ratio of reference significantly decreased P2X4R/P-P38expression of Radix curcumae extract high dose group, Gabapentin group in L4-L6the spinal cord (P<0.01or P<0.05).Conclutions:1. Study on the mechanism of neuropathic pain CCI rats:1.1The neuropathic pain induced by CCI rats can lead to:the body mass in CCI model rats decreased significantly, TWL、MWT value reduced.1.2The neuropathic pain induced by CCI rats can cause the rats model of peripheral blood BDNF, IL-1β concentration increased significantly, the high expression of BDNF in dorsal horn of spinal cord, and increase the expression of P2X4R P-P38.2Intervening effects of turmeric extract on neuropathic pain:2.1From the observation of clinical curative effect:14days of administration, Radix curcumae extract body weight of rats in high dose group was higher than other rats; and can prolong the CCI rat model of TWL, MWT significantly, which has the significant role of neuropathic pain.The improvement of high dose of Radix curcumae extract on neuropathic pain by reducing the peripheral blood BDNF, IL-1β concentration, reduce the expression of P2X4R, P-P38in the spinal cord; therefore, the possible mechanism is related with P2X4R/BDNF pathway.
