Inhibitory Effect Of Zhuanggu Analgesic Capsules On VEGFR Signaling Pathway In Nude Mice With Highly Metastatic Breast Cancer

Abstract:Objective:To discuss the inhibitory action and the mechanism of Zhuanggu Zhengtong capsules (ZZC) on transplanted tumor in nude mice bearing breast cancer.Methods:By built a model of metastasis in nude mice bearing breast cancer and32nude mice were randomly divided into four groups:the ZZC treatment group, the ZZC preventative group, the ShenYi capsules (SYC) control group and the model group. Give the medicine to the mice according to the dosage regimen and have a dynamic observe in the growing state of the transplanted tumor in every group. Meanwhile, we need to record the growing condition of the nude mice, get the weight and the weight changes of them, measure the volume of the transplanted tumor, and then plot a growth curve. After14days the mice would be executed, the tumors’weight will be measured, the inhibition rates will be calculated. Then we plan to determine the differences of the VEGF’s and MMP-9’s expression in tumor of different groups by immunohistochemistry.Result:The ZZC can obviously inhibit the growth of the transplanted tumor of breast cancer. The tumors’weight of the ZZC treatment group, the ZZC preventative group, the SYC control group was lighter than it of model group, and it was statistical significance observed (P＜.01) There was no significant difference in the weight of the tumor between ZZC treatment group and the SYC control group (P>0.05). The VEGF’s and MMP-9’s expression in tumor was markedly reduced of the ZZC treatment group, the ZZC preventative group, the SYC control group, and it is more obvious in the ZZC preventative group than two other groups which have no remarkable differences between them.Conclusion:1.The ZZC can obviously inhibit the growth of the transplanted tumor of metastasis breast cancer in human-nude mice.2.One of the mechanism of the ZZC’s inhibitory action is that the ZZC can suppress the endothelial cells’proliferation by reduce the VEGF’s and the endothelial cells’migration by reduce the MMP-9’s expression in VEGFR signal transduction of the tumor tissue, which result in the inhibition of the angiogenesis.
