Objective To observe the expression of miRNA-451in ischemic postconditioningattenuates lung injury induced by lung ischemia-reperfusion in rats and investigate thepossible mechanism, which sought to elucidate the protection mechanism of ischemicpostconditioning on miRNA regulation level.Methods Twenty-four SD rats were randomly divided into three groups (n=8in eachgroup): sham-operated group (sham), ischemia reperfusion group (I/R), ischemicpostconditioning group (IPO).Using real-time fluorescent quantitative PCR (RT-qPCR)to measure miRNA-451and Western blot detection p-AKT protein expression. Lungapoptosis was determined by TdT-mediated dUTP nick end labeling (TUNEL)methods.Results Compared with the sham group, miRNA-451expression in rat lung tissue inI/R and IPO group significantly higher (P<0.05), p-AKT expression was significantlydecreased (P<0.05); There are a large number of apoptotic cells in I/R group lungtissue, a small amount of cell apoptosis in IPO group and rare cell apoptosis in shamgroup.Conclusion IPO reduces the lung injury in rats induced by lung ischemia reperfusion,and its mechanism is possibly through down regulation of miRNA-451may beinvolved in AKT signaling pathway....
