Cervical cancer is second most common gynecological cancer just after breast cancer. In1995, International Association for Research on Cancer (IARC) identified human papillomavirus (HPV) type16and18as highly carcinogenic. Further research indicated that persistent HPV infection is a necessary cause of cervical cancer and type31,33,35,45,51,52,58,59are also highly carcinogenic. The detection rate of different HPV types differs by regions. In China, especially in the southeastern region the detection rate of HPV type58is high and even higher than type18in some areas, only after type16.Human papillomavirus is a double-stranded circular DNA virus with about200subtypes. It consists of6early proteins (E1, E2, E4, E5, E6, E7) and two late proteins (L1, L2), among which the E1protein is the most conserved protein in the entire HPV family. El protein plays an important role in the startup of viral DNA replication and affects the host cell growth cycle by reacting with E2protein. Given the high incidence of HPV58in China and the high conservativity of El protein, identification of linear B cell epitopes will contribute to the development of vaccines and antibody drugs.In this study, we expressed and purified the recombinant HPV58El-N and El-C proteins and immunized3male New Zealand white rabbits respectively to obtain anti-serum.7018mer peptides overlapping each other by9residues covering the entire El-N and El-C proteins were synthesized using the improved bio-synthetic peptide method and then detected through western-blot using the prepared anti-serum.24main B cell epitopes and5dominant epitopes(DETADDSGTDLIEFIDDSVQSAE TTQA, CTHRKRKIIELEDSGYGN, MSNISDVQGTTPEWIDRL, TVLQHSFNDDI FDLSEMI, EFNNPFPFDANGNPVYKI).This study lays the foundation for minimal motif mapping of linear B cell epitopes of HPV58El protein and will further contribute to the development of vaccines and antibody drugs.
