Lung cancer is one of the most common malignant tumors and it is the world’s highest incidence and mortality of cancer. It is a complex disease caused by a series of factors. Lung cancer is currently considered to be mainly caused by genetic polymorphisms and environmental factors.GWAS studies indicate that BAT3gene is one of the candidate lung cancer susceptibility genes. BAT3has been shown to be required for p53acetylation. BAT3also involves in apoptosis by interacting with the apoptosis-inducing factor (AIF), elongation factor XEF1AO and/or anti-apoptotic protein Bcl-2. In addition, BAT3acts as a TGF-P receptor-interacting protein and regulates TGF-β signaling.In order to study the association between BAT3gene polymorphisms and lung cancer susceptibility in Chinese Han population, we genotyped11tag SNPs of BAT gene among784cases and782age-and sex-matched cancer-free controls to screen for any potential SNPs associated with the disease. The results showed that BAT3-rs1077394GA and GG genotypes have protective effect in the female (adjusted OR=0.63,95%CI=0.42-0.95,P=0.030; adjusted OR=0.49,95%CI=0.25-0.93, P=0.030); BAT3-rs1077393GA and AA genotypes have significant protective effect in those over60years old (adjusted OR=0.69,95%CI=0.48-0.99, P=0.043; OR=0.69,95%CI=0.28-0.87, P=0.014). BAT3-rs2242656GA genotype has a significant protective effect in the people less than60years old and smokers (adjusted OR=0.64,95%CI=0.44-0.92, P=0.017; adjusted OR=0.58,95%CI=0.41-0.82, P=0.002).Haplotype analysis showed that rs1077394, rs3130047and rs1077393in BAT3gene can constitute a haplotype, and the haplotype ’GGA’ can significantly reduce the risk of lung cancer in the population less than60years old, female and non-smokers(OR=0.73,95%CI=0.55-0.95, P=0.022; OR=0.62,95%CI=0.45-0.85, P=0.003; OR=0.71,95%CI=0.56-0.9,P=0.004).The study indicated that the BAT3gene polymorphisms are associated with lung cancer susceptibility.
