Diabetes is a metabolic disease which has hyperglycemia characteristics. It is one of the ubiquitous chronic and systemic diseases. It is primarily induced by consolidated conditions including genetic factors, environmental factors, as well as other physical and chemical factors or emotional factors. However, its specific pathogenesis is not very clear yet. Although clinical diabetes drugs--chinese medicine, western medicine, chemical medicine, etc.--have some hypoglycemic effect on treating and improving some symptoms of diabetes, there are shortcomings such as side effects, unstable efficacy, expensive and so on. Moreover, such medication would cause further damage on kidneys, liver, etc. In recent years, some natural hypoglycemic drugs draw people’s attention due to safety, effectivity, little side effect, low price, etc. Ligustrum lucidum Ait contains terpenes, polysaccharides and a variety of chemical composition of phospholipids, volatile oils, flavonoids and so on, which have multiple functions in liver protection, anti-inflammatory, improvement of immune, anti-aging, anti-oxidation, etc. In this study, a mouse model of diabetes were caused by alloxan. We research effects of hypoglycemic activity and oxi dative stress level of raw material from L. lucidum and its wine broiled production in different extract parts. The main content is as follows:1. Firstly, according to the Chinese Pharmacopoeia and the relevant literature, we prepared wine broiled production of L. lucidum. We picked plump fruits which have consistent color, then washed and dried them. Wine broiled production was obtained by following steps:1)measure500g fruits and100ml rice wine accurately,2) mix and dilute with50%wine,3) incubate2h,,4)steam2h at100KPa,5)dry in the shade at room temperature and storeas an experimental material.2. Preparation of different extracts of raw material from L. lucidum and its wine broiled production. Total extract was obtained by refluxing with75%methanol, then its production was filtered, concentrated an,d freeze-dried. Before extract, the powder was dissolved in ddH2O, and then extracted respectively with ethyl acetate and n-butanol. Its production was concentrated and freeze-dried. Finally, we obtain methanol extract, ethyl acetate extract and n-butanol extract of raw material from Ligustrum lucidum Ait and its wine broiled production. 3. Study about hypoglycemic effects of different extracts of raw material from Ligustrum lucidum Ait and its wine broiled production. Clean grade Kunming male mice which was about four to six old ages and weighed18-22g were randomly divided into normal control group, diabetes model group, positive drug group, high and low dose group of methanol extracts of raw material, high and low dose group of methanol extracts of wine broiled production, high and low dose group of ethyl acetate extracts of raw material, high and low dose group of ethyl acetate extracts of wine broiled production high and low dose group of n-butanol extracts of raw material, high and low dose group of n-butanol extracts of wine broiled production with10mice in each group. After adaptively feeding for4d, we measured their weight and blood sugar. Except the normal control group, all of the groups were treated with alloxan solution200mg/kg· BW within a single intraperitoneal injection of0.2mL as diabetes model, and the diabetes model group injected with the same volume of normal saline. During the experiment, all the groups were given enough food and water freely without any intervention, however, not treated with other auxiliary hypoglycemic drugs and food. Methanol extracts of raw material, methanol extracts of wine broiled production, methanol extracts of wine broiled production, ethyl acetate extracts of raw material, ethyl acetate extracts of wine broiled production, n-butanol extracts of raw material, n-butanol extracts of wine broiled production were fed to the corresponding group with corresponding concentrations. Positive drug group was given an equal volume of metformin solution, normal control group and diabetic group were fed with an equal volume of saline regularly. Blood sugar were measured every week. Weight, water intake and food intake were, measured every dayfor3weeks. Oral glucose tolerance test(OGTT)was performed at the end of the third week, mice were killed for accurately measuring of the weight of stress indicator organs (liver, spleen, renal), measuring of the antioxidant enzymes activity in the liver tissue (T-SOD), lipid malondialdehyde (MDA), glutathione (GSH-PX) activity levels of hexokinase (HK), pyruvate kinase (PK) activity and glycogen content. Data was analyzed using SPSS software. According to the protocols of the kit, studies on the hypoglycemic effect of different extracts of raw material from L. lucidum and its wine broiled production. using biochemical analysis methods provide a theoretical basis for the further application and development of medicine, health production from L. lucidum.4. The results showed that:different extracts of raw material from L. lucidum and its wine broiled production can reduce blood glucose levels in diabetic mice, relieve symptoms of decreased weight, increased drinking, increased urination, and increased diet resulted from diabetes; different extracts of raw material from L. lucidum and its wine broiled production can relieve liver, spleen, kidney tissue hypertrophy and lesions in diabetic mice; different extracts of raw material from L. lucidum and its wine broiled production have free radical scavenging effect. They also can improve the body tissue’s diabetic mice antioxidant enzymes (T-SOD) activity and GSH-Px content, while they decrease MDA content in diabetic mice. This paper systematically studied therapeutic effect of diabetes with different extracts of raw material from L. lucidum and its wine broiled production. The research results can provide research data for depth research of hypoglycemic effect with different extracts of raw material from L. lucidum and its wine broiled production.
