Objective:Acanthopanax Senticosus is a famous Chinese traditional medicine, which belongs to the family Araliaceae. Modern pharmacology has demonstrated that the root and stem of Acanthopanax Senticosus has important activities such as replenishing vital energy, nourishing kidney-yin, tranquilizing mind and anti-fatigue. Acanthopanax root tablet is made of extractum acanthopanacis senticosi, which be put into widespread use in the clinic at present. There are some disadvantages to acanthopanax root tablet, such as large doses, poor compliance and so on. To reduce the doses, the topic has been studied that the preparation technology of the total eleutherosides tablet, and also the quality of the total eleutherosides tablet was discussed.Methods:Use HPLC method to measure the content of syringin, eleutheroside E and isofraxidin in Acanthopanax Senticosus. First, make sure that the menstruum is extracted, then use the orthogonal design method to investigate the influence that menstruum dosage, the extracting times and extracting time have on the extraction technology, eventually, nail down the optimal extraction technology. Compare the content of syringin, eleutheroside E and isofraxidin from various habitats in Acanthopanax Senticosus according to the optimal extraction technology.Use the absorption quantity and resolution of syringing, eleutheroside E and isofraxidin as the index of preparation of total eleutherosides to study the absorptive characteristics and elutive parameters of the process.A series of chromatographic and spectroscopic procedures were used to separate the total eleutherosides and the structures were elucidated on the basis of the physicochemical property and spectral features.Use headspace GC to determine residues in radix notoginseng extraction.Taking ginseng stem leaf saponin as positive comparison, compare the effect of total eleutherosides, acanthopanax root tablet and extractum acanthopanacis senticosi on the swimming time in the normal mice and the normal loaded mice, on the times of the spontaneous motion of the mice, on the sleep time in mice induced under pentobarbital sodium (threshold dosage), and on the latency of time and the time of sleep in mice induced by pentobarbital (threshold dosage).Faces, hardness, disintegration time, friability and dissolution of the tablets as the primary screening indexes to study the formula and procedure of total eleutherosides tablet.The weight difference, disintegration time and friability of the tablets were tested, and the contents of the effective ingredients in the tablets were determined by HPLC and ultraviolet spectrophotometry. Results:Optimal condition for extraction technology is as follows:extract3times and eachtime for2hours with10times the amount of50%ethanol. Acanthopanax Senticosus produced in Heilongjiang has higher quality. The absorption and isolation effect of AB-8type resin was much better. The optimalconditions were as:the concentration of sample solution was0.5g·mL-1, the adsorption flow rate was2BV·h-1the maximum loading amount was5BV, the eluent was30%ethanol, the dosage was11BV and the elution flow rate was1BV·h-1. A total of nine known compounds are isolated from total eleutherosides and indentified aseleutheroside E(1), eleutheroside B1(2), Syringaresinol-4-O-β-D-glucoside(3), glucosyringic acid(4), sinapaldehyde glucoside(5), daucosterol(6), syringing(7), isofraxidin(8) and β-ecdysterone(9). Dot not detect N-hexane, benzene, o-xylene and styrene in the total eleutherosides.The total eleutherosides could significantly postpone the sustained swimming time in thenormal mice and the normal loaded mice, reduce mice’ the times of the spontaneous motion, increase the sleep rate of mice on the the sub-threshold dose of pentobarbital sodium and prolong the sleep time of mice on the threshold dose of pentobarbital sodium.lt shows that the total eleutherosides has a notable anti-fatigue and sedative effect. The optimum formulation of total eleutherosides tablet was composed of major medicine0.06g, MCC0.03g, dextrin0.03g, CMC-Na0.012g and magnesium stearate0.36mg.The contents of syringing, eleutheroside E, isofraxidin and total eleutherosides should not be lower0.7mg/tablet,1.7mg/tablet,0.3mg/tablet and38.9mg/tablet. Conclusions:The extraction process of the effective part and total eleutherosides fromAcanthopanax Senticosus with the major effective ingredients all adopted as evaluation indexes, and explore the chemical constituents of total eleutherosides and the preparation technology of total eleutherosides. The total eleutherosides had high purity, twhich were approximately the same effect as the market preparations in terms of the contents of crude drugs. The quality of total eleutherosides tablet was controlled, and it had the advantage of small doses. So it will have a certain market value.
