Objective:To investigate the single nucleotide polymorphisms (SNPs) of rs13129697/rs7442295within SLC2A9gene in Han Chinese Males, and to examine the association of two SNPs within this gene with diabetes mellitus (DM), hyperuricemia (HUA), diabetes mellitus complicated hyperuricemia.Methods:A total of285DM subjects,300HUA subjects,198DM complicated HUA subjects and550healthy controls were recruited in this study from gout clinic, endocrinology clinic and ward, physical examination center in the affiliated hospital of medical college of Qingdao university. Variants rs13129697, rs7442295within SLC2A9were genotyped by using TaqMan-MGB Duplex real-time PCR. The differences of genotypes, and the genotype and allele frequencies for Hardy-Weinberg equilibrium proportions were analyzed by χ2test, Adjusted odds ratios and95%confidence interval were calculated by multiple logistic regression analysis.Results:The frequencies of GG, GT and TT genotypes of rs13129697in controls were21.6%,56.4%,22.0%respectibely, while they were22.4%,51.6%,26.0%in DM patients respectibely, in HUA patients were14.3%,54.0%,31.7%, in DM complicated HUA patients were15.7%,54.5%,29.8%. We observed significantly statistical difference in distribution genotype frequencies of rs13129697between control group and hyperuricemia group (X2=11.225, P<0.001) as well as diabetes mellitus complicated hyperuricemia group (X2=5.536, P<0.05). Logistic regression in an age, triglyceride and cholesterol model suggested that people carrying GT, GG allele had lower risk of HUA(OR=0.615,95%CI:0.432-0.877,P=0.007;OR=0.447,95%CI:0.271-0.736,P=0.002) as well as DM complicated HUA (OR=0.578,95%CI:0.360-0.923,P=0.022;OR=0.393,95%CI:0.208-0.740, P=0.004), and there were statistical significance. The frequencies of AA and AG genotypes of rs7442295in controls were96.2%and3.8%respectibely, while they were96.5%and3.5%in DM patients respectibely, in HUA patients were96.0%and4.0%, in DM complicated HUA patients were96.0%and4.0%, the GG genotypes was completely absent among the study population. However, Genotype frequency distribution difference between these four groups was not statistically significant (P>0.05).Conclusion:The SNP rs13129697within SLC2A9gene is associated with hyperuricemia and diabetes mellitus complicated hyperuricemia in Han Chinese males. The allele G is protective for human. The present data suggests the rs7442295SNP of SLC2A9gene have no association with diabetes mellitus,hyperuricemia and diabetes mellitus complicated hyperuricemia.
