Objectives: By studying the changes for mitochondrial fusionprotein Mfn-1and fission protein Drp-1gene expression after exhaustive exercise inrat cerebral cortex, and the relationship between cell apoptosis. Preliminary discussionon the exhaustive exercise resulted mechanism of cell apoptosis in cerebral cortex.Research methods: choose180~220g of adult male rats40, the rats were randomlydivided into5groups: the blank group, immediately after exercise group,12hoursafter exercise group,24hours after exercise group,48hours after exercise group andestablish the model. The blank group without any intervention treatment, immediatelyafter exhaustive exercise were immediately after exercise group, exhaustive samplingof12hours,24hours,48hours after exercise respectively for12hours after exercisegroup,24hours after exercise group,48hours after exercise group. Expression genesBax and Caspase-3observation of cell apoptosis in cerebral cortex in each group byimmunohistochemical; The quantity mRNA expression of Mfn-1and Drp-1to detect incerebral cortex cells by real-time fluorescence quantit-ative PCR technique; Theexpression of Mfn-1and Drp-1in cerebral cortex cells by Western blot protein.Research results:(1) Immunohistochemistry: Induced apoptosis regulatory genes Baxand Caspase-3expression increased after exhaustive swimming exercise, the proteinexpression positive level rate of Bax, Caspase-3: There are obvious differences in theprotein expression level of blank group and the other groups (P<0.01); Had significantdifference protein expression level between immediately after exercise group and theother groups (P<0.01);12hours after exercise group protein expression is the highesthad significant difference protein expression level between and the other groups(P<0.01).Had significant difference protein expression level between24hours afterexercise group and the other groups (P<0.01); Had significant difference proteinexpression level between48hours after exercise group and the other groups (P<0.01).(2)RT-PCR:expression of mRNA mitochondrial Drp-1, Mfn-1results showed anupward trend with immunohistochemistry in rat brain cortex after Exhaustiveswimming exercise. The blank group Drp-1, Mfn-1mRNA expression is obviouslydifferent between the other groups (P <0.01); The blank group Drp-1, Mfn-1mRNAexpression is obviously different between the other groups (P <0.01); immediatelyafter exercise group Drp-1, Mfn-1mRNA expression is obviously different between the other groups (P<0.01);12hours after exercise group Drp-1, Mfn-1mRNAexpression is obviously different between the other groups (P<0.01);24hours afterexercise group Drp-1, Mfn-1mRNA expression is the highest obviously differentbetween the other groups (P<0.01);48hours after exercise group Drp-1, Mfn-1mRNAexpression is obviously different between the other groups (P <0.01), The groups wasstatistically significant data.(3) Western Blot: Expression of Drp-1gene protein expression is minimum inblank group,had significant difference between the other groups (P<0.01); Expressionof Drp-1gene protein expression is rise in immediately after exercise group,hadsignificant difference between the other groups (P<0.01); Expression of Drp-1geneprotein expression is rise in immediately after exercise group and12hours afterexercise group, had significant difference between the other groups (P<0.01);Expression of Drp-1gene protein expression is rise in24hours after exercise groupand48hours after exercise group, had significant difference between the other groups(P<0.01). Expression of Mfn-1gene protein blank group are the same in blank groupand immediately after exercise group,had significant difference with the other groups(P<0.01); Expression of Mfn-1gene protein expression of12hours after exercisegroup and24hours after exercise group are the same, had significant difference withthe other groups (P<0.01); Expression of Mfn-1gene protein expression of48hoursafter exercise is the highest,had significant difference with the other groups (P<0.01).Conclusion: Gene expression changes for rat cortex cell are the same betweenmitochondrial fusion and fission proteins and cell apoptosis after exhaustive exercise,exhaustive exercise could be the change in expression of mitochondrial fusion fissiongene of apoptosis.
