| ObjectiveTo observe the infection of PI3K/AKT signal transductionpathway inhibitor LY294002,cisplatin about TCA8113TCA8113/CDDP cells’s proliferation, and to study on therelationship between signaling transduction pathway and cisplatin drug resistence.Methods1.Gradually increasing the concentration of CDDP added to the cells TCA8113and addedcontinually, through this methods we gained the TCA8113/CDDP cell lines.The TCA8113celllines wes theated with0.3μg/mLCDDP for two days,and then change for the normal culturemedium.when the cells was subcultured normally,adding CDDP twice.The cell lines wascultured for one week before using the second concentration,which is one time as the startingconcentrion.2.We treated the TCA8113and TCA8113/CDDP cell lines with different concentration(0μmol/l,5μmol/l,10μmol/l,20μmol/l,40μmol/l)of LY294002for24and48hours. Thecombined inhibition rate was calculated by the concentrion of cisplatin IC50and LY294002for24h.The inhibition rate of are determined by mtt assay.3.The experiment was devided into the.blank group the control group and the experimentalgroup.The blank group were added the culture medium,the control group were added thecisplatin with IC50concentrion,the experimental were added with different concentrion ofLY294002and cisplatin IC50.All the group were cultured for24hours,and extrating the protein.The expression results of PI3K, p-AKT, AKT were analysed by western blot.Results1.We gained the Tca8113/CDDP cell lines and the cells’ CDDP drug resistence ability was7.69.2.The LY294002and cisplatin inhibition rate of TCA8113and TCA8113/CDDP cells aretime and dose-dependent, the combination of LY294002and CDDP showed a synergistic effecton the two cell lines’ growth inhibition.3.The band line of p-AKT, AKT and PI3K were obviousely thin. PI3K, AKT and p-AKTexpression of TCA8113/CDDP are much more than that of TCA8113.Conclusion1.LY294002could inhibite the growth of TCA8113and TCA8113/CDDP cell lines,and theinhibition rate is time and concentrition-dependent.2.LY294002could induce TCA8113and TCA8113/CDDP sensitivity to CDDP... |
