Objective: To study the affects of Streptococcus sanguis bacteriocins on the growth curve.To discuss the affects of Streptococcus sanguis bacteriocins on the intracellular free calciumconcentration of Porphyromonas gingivalis.Methods:1.The standard strain of Streptococcus sanguis ATCC10556was recovered andcultured and then intracellular proteins of Streptococcus sanguis(Streptococcus sanguisbacteriocin) was extracted by centrifugation and broken.2.Using trace liquid times dilution method was developed for the determination ofstreptococcus sanguis bacteriocins of P.g minimum bacteriostatic concentration (MIC).3.Choose different concentrations of streptococcus sanguis bacteriocins affect P.g,respectively at0,4,8,12,16,20,24and48h to measure the determination of groups ofbacteria suspension at590nm wavelength optical density (OD) value and then drew the growthcurve with time as abscissa and OD value as ordinate. To study the effect of Streptococcussanguis bacteriocin on growth curve of Porphyromonas gingivalis4.Use Fluo-3/AM load P.g intracellular calcium ions, use the Laser confocalmicroscope observation of Ca2+fluorescence intensity changes.Results:1.Streptococcus sanguis bacteriocins has a significant inhibitory effect of P.g, the MICis0.0625g/L.2.After the Streptococcus sanguis bacteriocins works, P.g intracellular calciumion concentration decreased significantly.2.The growth curve of P.g has a significant change occurred in8~16h, logarithmicphase is delayed for eight hours.3.The intracellular calcium ion concentration of.P.g significantly reduced, statisticallysignificant difference compared with the control group(P<0.01). Conclusion:1.Streptococcus sanguis bacteriocins for P.g has obvious inhibitionbacterium.Within the scope of this study, with the increase of streptococcus sanguis bacteriocinsits inhibition enhanced gradually.2.Streptococcus sanguis bacteriocins can cause P.g cell membrane permeabilityenhanced calcium bacterium.
