| In this experiment two varieties of Zingiber offcinai Roscoe.that named Liujiang No.1 and Ludajiang were used as the materials.The system of tissue culture, micro-ginger induction and agrobacterium-mediated transformation of Zingiber offcinai Roscoe. was established and some GUS transgenic plants of Zingiber offcinai Roscoe. had been obtained successfulIy.The results offer significant referenced datum for the future research work.The results of study were included in the following:1 0.1%HgCl2 12min+70% CH3CH2OH 1min was suitable to sterilited the shoot-tip of Liujiang No.1,the contamination rate was 12.1%, the rate of buds differentiation was 73.5% and the average of induced bulblets was 2.46. That of Ludajiang was 15.6%,79.3% and 2.58.2 There were different frequencies of buds induction and differentiation with different genotype varieties,the frequency of buds differentiation with Liujiang No.l was 97.5%,the average of induced bulblets was 2.55. That of Ludajiang was 84.17% and 2.16.3 The pathway of adventitious bud differentiation may become the main pathway on tissue culture of the Zingiber offcinai Roscoe. The differentiation rate of callus produced by the pathway of de-differentiation was low, which was disadvantage to tissue culture of Zingiber offcinai Roscoe.4 The buds and stems were the suitable materials for tissue culture of Zingiber offcinai Roscoe., which showed the high differentiation rate of adventitious buds.The buds differentiation rate of callus,which induced by buds ,was higher comparatively.5 On the buds callus culture,the optimal medium of the buds differentiation was MS+6-BA1.0mg/L+2,4-D3.0mg/L.To Liujiang No.1,the rate of callus induction was 84.6%,the rate of buds differentiation was 88.6%,and the average No.of buds... |
