Populus alba Linn. is one of Salicaceae Linn., and Ulmus pumila Linn. is one of Ulmaceae Ulmus Linn. Wide hybridization of them which regarded Populus alba Linn. as female parent , and Ulmus pumila Linn, as male parent began in 1979. Crossing Salicaceae with Ulmaceae was made by the use of pollen inducement and ovary culture technology in 2004 and 2005. At the same time, Paternity test of 1979's hybrid offspring of Populus alba Linn. and Ulmus pumila Linn. by means of the technology of morphology marker, cytology marker, hybrid test and AFLP marker. Mean while, The experiment of pollination,the growth and the propagating method of generation was also carried out. By these experiments, the major results and conclusions are described as follows.At first, there is a misogamy in meiosis in the hybrids .But the cross difficulty can be conquered by the technology of pollen inducement and ovary culture.Secondly, the results of paternity test show that filial generation (It is called P.×yinyu as follow) is more similar to female parent in phenotype. But P. × yinyu blooms early, its twigs are slim and fragile, no hair on its back of green leaf, and it is androgynous , which all of them are similar to Ulmus pumila Linn..There are 38 chromosomes in P. yiny as same to the poplar. I presume it does not belong to the union of the sperm and egg. P. × yinyu's selfing shows that phenotype of its F2 generation is similar to Populus alba Linn, but most of them have either more or least characters of Ulmus pumila Linn. AFLP markers of parents and filial generation indicate that filial generation does contain of Ulmus pumila Linn..Thirdly, study on value breeding of P.× yinyu shows that pollens of P. × yinyu is vital by tests of dyeing, tissue culture, crossing, but there are about thirty percent pollen tubes that are tortuose, forficate in tissue culture, which is likely to produce cross incompatible or crossing failure. But sterility is not obvious in selfing. Another, the pollens' amount of P.× yinyu are few.Pistil of P. × yinyu is also pregnant by tests of (P. × yinyu) ×Populus alba var. Pyramidalis Bunge etc.. There is different affinity of different cross combination.Fourthly of field test of P. × yinyu. The growth rate of P. × yinyu is common. The average height of 11 ages old P. × yinyu is 18.0m.
