Triticale is a new man-made crop,which comes from triticum and secale hybridization,It has a good adaptability to resistance of worse environments. The main studies of triticale focus on acid resistance and barren endurance ect,but the report of resistance of drought is seldom. In the global water resources become increasingly tense occasion triticale research molecular mechanisms of drought on new varieties of triticale cultivated drought extremely far-reaching significance.The experiments used against drought stronger TriticaleH01-4 with Spring 2nd for comparison with 30%PEG-6000 Gaoshen solution drought simulation environment for seedlings to address drought, respectively 0h, 24h, 48h, 72h and 48h of water, stress 48h sampling, three separate studies the identification of drought, including leaves relative water content (RWC),proline (Pro) content , Coleoptile lengh,main radical lengh; SDS-PAGE study of the protein changes drought stress, and provide a theoretical basis for the differences display, thus showing DDRT-PCR. Research content and results are as follows :1.30% of concentrations of stress triticale and spring 2nd each contributed significant level, the experiments used stress agent for 30%PEG-6000 concentration.2.Water stress obviously restrain growth of coleoptile and main radical. Coleoptile length of view, with the extension of stress time, Coleoptile length of triticale the relatively changes rate is higher than spring 2nd. Main radicle lengh of triticale stress smaller show that the drought of triticale stronger.3.Proline capacity of accumulation is related with drought stress.With the extension of drought stress, accumulation of high levels of triticale proline has strong ability of hold water, the leaves are able to maintain relatively high water content.4.Stress through different time under the leaves solubility protein SDS-PAGE electrophoresis, the results show that : 5 belt of triticale H01-4 changed, with the extension of stress, disappearance of a belt, content of a belt gather head.and giving out three new protein belt .Spring 2nd belt only two occurred in the change of content.5.There will be guided by three anchors primer, which are composed of eight randomly drawn from 24 different combinations for each of the two triticale and wheat crops in comparison and 24h and 72h stress under the leaves for DDRT-PCR analysis showed different from primer combination of different crops produce total of 104 different fragments.
