| The disease caused by Whitefly-transmitted Geminivirus (WTG), a type member of Geminivirus, is an important one to many economic crops in the world. It has caused serious loss to the crop yield of 39 countries. These years it has broken out in some areas of Yunnan, Hainan ,Guangdong and Guangxi provinces. However, there aren't interrelated reports to prove the existence of WTG in Fujian province. The survey in fields showed that the Malvastrum coromandelianum disease symptom caused by WTG was typical. We identified the geminivirus in Malvastrum coromandelianum from the molecular level by designing the primers, PCR, cloning and sequencing.Two virus samples were collected from Malvastrum coromandelianum and these samples were tested by PCR. The results showed that a specific band about 500 bp was amplified from the samples Fz1 and Fs1 with degenerate primers PA and PB. The results showed that the both samples were infected by geminiviruses. Two fragments from Malvastrum coromandelianum samples were cloned and sequenced. Sequence analysis of PCR products showed DNA fragments from Fzl and Fsl have 96% sequence homology each other. The result showed that the virus isolates from Fzl and Fsl belong to the same virus. On the basis of these data, complete genomic structure of virus infected Fzl was studied.The complete nucleotide sequence of Fzl DNA-A was determined, it contains 2741 nucleotides, having typical genomic organization of a begomovirus, encoding six ORFs with two ORFs [AV1(CP) and AV2] in virion-sense DNA and four ORFs (AC1-AC4) in complementary-sense DNA Similarity researches showed that Fzl DNA-A has the highest sequence identity (86%) with those of Papaya culr leaf Fujian virus (PaCuLGDV-[GD2], AJ558122) and Papaya culr leaf virus (PaCuLCNV, AY650283), while less than 83% identities are found when compared with other Begomoviruses.
