Cucumber Angular Leaf Spot, Cucumber Scab and Cucumber Anthracnose are the main diseases in the cucumber production. The occurrence of these diseases is very widespread all year around and seriously affects safety of vegetable production. Cucumber Pink Mold Rot and Cucumber Corynespora Target Leaf Spot have occurred recently in the North China. They spread fast. The diagnosis of diseases is the key measure to control disease. The regular diagnosis Methods have the disadvantages of depend on experience and diagnosis. To solve this problem, we aim to set up a series of fast, accurate and sensitive diagnosis methods for the detection of these five diseases.After research on the detection of cucumber diseases by ELISA, we found the methods to prepare the antigens of pathogenic fungi and bacteria, compared the characteristics and effect of two different methods (with fungi and bacteria) of antibody preparation. The quality control parameters of disease detection by ELISA were determined. The facts proved that the ELISA method is feasible in disease diagnosis. This provides the technique parameters for the application of the disease diagnosis kit.The pathogenic fungi was cultured in liquid culture medium. Which solves the problem that some mycelium of fungi can not grow well in solid medium and laid the groundwork for mass purification of mycelium proteins. The NA culture medium is used to culture the pathogenic bacteria, which is inactivated and centrifuged before it is used as antigen to prepare antibody. This method to prepare antigen has the advantage of simpleness and low cost, compared with the method to purify the targeted composition from pathogen. And the prepared antibody, with the titers over 20,000, can meet the demand of diagnosis. Four different coating buffers were selected to coat the antigen and their effects ondiagnostics were compared respectively. The result showed that the 0.05mol/L, pH9.6Carbonate coating buffer had the best effect and it can be applied to most diagnosticskits.
