Three different methods of extracting Platycodon platycodin (Sorbitic extraction, ultrasonic extraction, microfarad extraction) were carried out in this study, the results showed that ultrasonic extraction was saving time, no heating, saving energy, not destroying the effective ingredient, and more effective , so this method was adopted in this experiment.Twenty five different roots of two-year Platycodon colleted in 3 different periods from the mid-August to the early October were used in the method of weighing and comparing colors. The result shows that comparing colors had a high sensitivity and little error, and the result was much similar to former ones, so it was used in this study.Twenty five different roots were collected 3 different periods, which were one-year Platycodon collected in early October , two-year Platycodon collected in mid-August and dry roots collected in early October ,were used to determine platycodoside .The result showed that the platycodoside in the root of Platycodon was related to their lifespan. Two-year Platycodon collected in early October was the most platycodoside, two-year Platycodon collected in mid-August was less . and the one-year Platycodon collected in October was the least . There was differences of platycodoside in different idioplasmic sources of the Platycodon, the purple-flowered Platycodon had more platycodoside than the white-flowered one. 3 times of determining platycodoside in 22 kinds of purple-flowered Platycodon showed that the average content ranged from 2.630% to 3.517% and 3 kinds of white-flowered Platycodon ranged from 2.499% to 2.608%.The platycodoside contents in different organs showed that the order from high to low was flower, ovary, top of reed, root leather, below top of reed, taproot and stem.The analysis between platycodoside of the 25 kinds of different roots and shape and property showed that there was no obvious correlation in one-year Platycodon; obvious positive correlation to the height of the plant (r=0.402*) and negative correlation to the weight of dry root(r=-0.494*) in two-year Platycodon.The platycodoside in 25 different Platycodon were obviously correlated to the developed properties in the tissue of the leaves and there was no obvious correlation to the RAPD analysis tree.
