The studies on the orchid mycorrhizal fungi have attracted great attentions in recent years. The orchid mycorrhizal fungi live in the special niche within the orchid roots. The co-evolution occurs between them. The fungi rely on the orchid for nitrition and meanwhile play important roles in the growth and the resistance to pathogeny and diseases of the orchid. The pharmaceutical components mainly produced by the medicinal plants were also found in some orchid micorrhizal fungi.Pleione bulbolodioides belongs to Pleione in Orchidaceae. The flower is very c-olorful and beautiful,so many people are fond of it. Above all,it possess medical function and therefore have been applied for many years. The medicinal functions of it ma inly include treating scrofula and detoxifcation, curing carbuncle and furuncle, healing bite by snake or insect, abating phlegm, and diminishing fever. Recent pharmacology- ical studies have found that it have the more important functions of anticancer. This had attracted many experts to study on it.The studies on the methodology for isolating , identifying the micorrhizal fungi of Pleione bulbolodioides using morphological biology techniques, propagation of the orchid by symbiotic seed germination, the effect of the production of fermentaion of the single isolate on the tissue culture of it and infection the Dendrobium loddigesii by the part isolates have been carried out. To isolate the micorrhizal fungi, 75% alcohol and 0.1% HgCl2 were used for surface disinfection. The disinfention time of using HgCl2 was 110min. And the best time is 25min. Isolated fungi belong to 19 genera in 9 families, 8 orders, 5 classes and 3 subphylums.They sum to 45 strains. The dominant isolates were deuteromycotina and ascomycetes. Ceratocystis, Monilia, Phialophora and other 4 genera were firstly isolated from orchid. For the symbiotic seed germination on oatmeal agar(OMA) medium, 69% single isolates promoted the seed gemination. 37.9% single isolates had a than contrail's germination rate.The GN21, as compared to the controll and other isolates, had a more than 80% germination rate.
