Bacteriostatic action of 28 Chinese herbal medicine on Escherichia coli ATCC11411, Staphylococcus aureus ATCC26112, and Pseudomonas aeruginosa was studied experimently. In these medicines, the extracts of Sophoraflavscens Ait had the obvious action on these three bacteriostasis, then the bacteriostatic action and mechanism and its effect on colibacillosis in chiken were studied. The result showed that the extracts had the obvious action in bacteriostasis upon Gram-positive and Gram-negative bacteria and the bacteristatic spectrum was wide. Primarily analysis indicated that the alkaloid of Sophoraflavscens Ait is the principal material for bacteriostatic action. Drafted growth curves of bacteriostasis uncovered the E.coli 01 C84010 effected by drug went into decline phase straight, got across the normal logarithmic phase. Observation under scanning election microscope revealed that the E.coli 01 C84010 had been constricted and distorted, depressed on center and just like the shoe-shaped gold ingots. Observation under transmission electron microscope revealed that cytoplasm was concentrated, cell membrane was separated from cell wall, then cell wall was damaged, the contents were leaked out and the E.coli 01 C84010 were killed finally. Analysis by flow cytometry indicated that after effecting by drug for 4.5 hours, the count of bacteria in I phase increased and that in R phase reduced by 20%. SDS-PAGE found that the extracts restrained the expression of functional protein, the total protein contents dropped down to one seventh of original bacteria, some banks disappeared and the action became intensively with the time extended. The present study suggested that the bacteriostatic mechanism was Sophoraflavscens Ait inhibited the synthesis of proteins associated with growth and cleavage of bacteria, then blocked their division and development. Furthermore, the drug combined with the protein, made the cytoplasm pyknosis and killed the bacteria finally. The clinical trial of Sophoraflavscens Ait prove that the extracts have preferabal effect on the colibacillosis. The experiment of acute toxicity showed that the toxicity of Sophoraflavscens Ait was soft, so it had the practice potent of substituting antibiotic to become the new-style animal remedy.
