Preliminary Research On Tissue Culture Of Pelargonium Graveolens

Pelargonium graveolens, a perennial balmy plant, can release citronellal which is the pesticide to mosquito. As a new environment-protected high-tech plant, Pelargonium graveolens has been cultivated in thousands upon thousands families of many countries, such as Japan, Australia, America and Canada. The prospect of Pelargonium graveolens application is very extensive. The market benefit is obvious and the demand is increased day by day.In our study, the effects of different plants growth substances on induction of Pelargonium graveolens callus and organ genesis were investigated by tissue culture and rapid-reproducing technology. The main results as follows:1. Using the young leaves of Pelargonium graveolens, the effect of different kind, different concentration and different combination of plant growth substances on induction of callus is different. The optimal medium for induction of Pelargonium graveolens callus is MS+2,4-D 0.2 mg/L+6-BA0.5 mg/L+sucrose 30 g/L+agar 5 g/L, pH=5.8.2. The effect of light on induction of callus: In light or in dark, the life circle is all 30 days. The reproduction curve is shown "S" accorded with Logistic Growth Model including 3 stages: delay stage, exponential growth stage and quiescence stage. In light the callus grew quickly.3. The study of organ genesis and plant regeneration: Roots and shoots could be differentiated from explants, but the differentiating rate is low and so is livability. The induction of organ differentiation from callus is easy. The medium of shoot induction is MS+6-BA 1.0 mgL^-1+NAA 0.1 mgL^-1+sucrose 30 g/L+agar 5 g/L, pH5.8. The exercising seedling medium is MS+6-BA0.1 mgL^-1+GA0.1 mgL^-1+sucrose 30 g/L+ agar 5 g/L, pH5.8. The medium for tube seedling root-generation is 1/2MS+NAA 0.1 mgL^-1 sucrose 15 g/L+agar 5 g/L,pH5.8. The key steps of Pelargonium graveolens transplant rate is exercising seedling which could increase the resistance of tube seedling.According to the different months, the condition of culture could be adjusted. The management after transplantation is also important.
